Snakebite envenomation has been a long-standing global issue that is difficult to treat, largely owing the flawed nature of current immunoglobulin-based antivenom therapy and complexity snake venoms as sophisticated mixtures bioactive proteins peptides. Comprehensive characterisation venom compositions essential better understanding toxicity inform effective rationally designed antivenoms. Additionally, greater composition will likely unearth novel biologically active peptides have promising therapeutic or biotechnological applications. While bottom-up proteomic workflow main approach for cataloguing at toxin family level, it unable capture heterogeneity in form protein isoforms higher-order interactions are important driving but remain underexplored. This review aims highlight importance beyond primary sequence, post-translational modifications give rise different proteoforms myriad complexes venoms. We focus on top-down workflows identify further discuss alternative separation, instrumentation, data processing strategies may improve proteoform identification. The structural techniques implemented also discussed; we emphasise need complementary higher resolution bioanalytical such mass spectrometry-based approaches, X-ray crystallography cryogenic electron microscopy, elucidate poorly characterised tertiary quaternary structures. envisage expansion "toolbox" with proteomics high-resolution structure determination be pivotal advancing towards development improved biotechnology

Background Snakebite envenomation inflicts a high burden of mortality and morbidity in sub-Saharan Africa. Antivenoms are the mainstay therapy envenomation, there is an urgent need to develop antivenoms broad neutralizing efficacy for this region. The venoms used as immunogens manufacture snake normally selected considering their medical importance availability. Additionally, ability induce antibody responses with capability should be considered, issue that involves immunization scheme animal species being immunized. Methodology/Principal findings Using lethality neutralization assay mice, we compared intrageneric scope antisera generated by horses monospecific, bispecific/monogeneric, polyspecific/monogeneric formulated Bitis spp., Echis Dendroaspis spitting Naja spp. or non-spitting It was found raised all were able neutralize homologous and, single exception, heterologous congeneric (considering separately). In general, polyspecific spp, spp gave best profile against these genera. For venoms, no significant differences between bispecific antisera. A similar result obtained case except antiserum more effective N . melanoleuca nivea monospecific antiserum. Conclusions/Significance use alternative produce monogeneric wide coverage African snakes , (non-spitting) On other hand, immunogen composed venom nigricollis suitable antivenom These can design

Snake venoms are comprised of bioactive proteins and peptides that facilitate severe snakebite envenomation symptoms. A comprehensive understanding venom compositions the subtle heterogeneity therein is important. While bottom-up proteomics has been well-established approach to catalogue compositions, top-down emerged as a complementary strategy characterize at intact protein level. However, not widely implemented in snake field proteomics, with various emerging methods yet be developed for systems. Here, we have explored three main mass spectrometry methodologies proof-of-concept study selected three-finger toxin phospholipase

Snakes responsible for bites are rarely identified, resulting in a loss of information about snakebites from venomous species whose venom effects poorly understood. A prospective clinical study including patients bitten by snake was conducted Cameroon between 2019 and 2021 to evaluate the efficacy tolerability marketed polyvalent antivenom. Clinical presentation during first 3 days hospitalization recorded following standardized protocol. This ancillary aimed assess frequency different encountered describe symptoms formally identified species. Of 447 included study, 159 (35.6%) brought that caused bite specialist. Out these, 8 specimens could not be due poor condition, 19 were non-venomous species, 95 belonged Echis romani—formerly E. ocellatus—species. The remaining 37 2 Atheris squamigera, 12 Atractaspis spp., Bitis arietans, 11 Causus maculatus, 1 Dendroaspis jamesoni, Naja haje, N. katiensis, 5 melanoleuca complex, nigricollis. Symptoms, severity envenomation, post-treatment course described. Symptoms consistent with cases described literature, but some specific features highlighted.
