Stress triggers anticipatory physiological responses that promote survival, a phenomenon termed allostasis. However, the chronic activation of energy-dependent allostatic results in load, dysregulated state predicts functional decline, accelerates aging, and increases mortality humans. The energetic cost cellular basis for damaging effects load have not been defined. Here, by longitudinally profiling three unrelated primary human fibroblast lines across their lifespan, we find glucocorticoid exposure energy expenditure ~60%, along with metabolic shift from glycolysis to mitochondrial oxidative phosphorylation (OxPhos). This stress-induced hypermetabolism is linked mtDNA instability, non-linearly affects age-related cytokines secretion, aging based on DNA methylation clocks, telomere shortening rate, reduced lifespan. Pharmacologically normalizing OxPhos activity while further increasing exacerbates accelerated phenotype, pointing total as potential driver dynamics. Together, our findings define bioenergetic multi-omic recalibrations stress adaptation, underscoring increased interrelated features load.

Brain vascular health appears to be critical for preventing the development of amyotrophic lateral sclerosis (ALS) and slowing its progression. ALS patients often demonstrate cardiovascular risk factors commonly suffer from cerebrovascular disease, with evidence pathological alterations in their small cerebral blood vessels. Impaired brain has detrimental effects on motor neurons: endothelial growth factor levels are lowered ALS, which can compromise cell formation integrity blood–brain barrier. Abstract Ageing and chronic stress have been linked to reduced telomere length (TL) in mixed‐age groups. Whether response components are TL during the midlife‐to‐late adulthood transition remains unclear. Our study aimed synthesise evidence on relationship between psychological biological of middle‐aged older adults. We conducted a systematic review studies obtained from six databases (PubMed, CINAHL, EMBASE, PsycINFO, Web Science, Scopus) evaluated by two independent reviewers. Original research measuring human individuals were included. From an initial pool 614 studies, 15 included ( n = 9446 participants). Synthesis showed that higher (i.e., global perceived or within specific life domain cognitive appraisal social‐evaluative stressors) shorter TL, specifically women under major stressors. For response, cortisol, dehydroepiandrosterone sulphate IGF‐1/cortisol imbalance, IL‐6, MCP‐1, blood pressure, heart rate presented significant association with but this depended stressors context (manipulated vs. non‐manipulated conditions). This comprehensive mainly those stressor stress‐induced conditions.
