Abstract Genome-wide association study has identified fruitful variants impacting heritable traits. Nevertheless, identifying critical genes underlying those significant been a great task. Transcriptome-wide (TWAS) is an instrumental post-analysis to detect gene-trait associations focusing on modeling transcription-level regulations, which made numerous progresses in recent years. Leveraging from expression quantitative loci (eQTL) regulation information, TWAS advantages detecting functioning regulated by disease-associated variants, thus providing insight into mechanisms of diseases and other phenotypes. Considering its vast potential, this review article comprehensively summarizes TWAS, including the methodology, applications available resources.

Mulberry (Morus alba L.) is a flowering tree traditionally used in Chinese herbal medicine. leaf flavonoids (MLFs) have been reported to exert important anti-inflammatory and antioxidant properties. The purpose of this study was select the MLF with best antioxidative activities from MLFs eluted by different ethanol concentrations (30%, 50%, 75%) explore its pharmacological Three types inhibited production nitric oxide (NO), prostaglandin E2 (PGE2), inducible synthase (iNOS), cyclooxygenase-2 (COX-2), inflammatory cytokines lipopolysaccharide (LPS)-induced RAW 264.7 cells. All boosted capacity decreasing reactive oxygen species (ROS) scavenging 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals improving metal ion chelating activity reducing power. results revealed that 30% exhibited activities. A nontargeted metabolomic analysis analyze 24 differential between MLFs. Quercetin, kaempferol, their derivatives 30%MLF were more abundant than other two Furthermore, we evaluated dextran sodium sulfate (DSS)-induced ulcerative colitis (UC) mice. could alleviate clinical symptoms, reduce secretion cytokines, inhibit activation pathway DSS-induced This will provide valuable information for development as functional food.

Abstract Ulcerative colitis (UC) is a recurring autoimmune disorder characterized by persistent inflammation in the mucosal lining of lower part large intestine. Conventional treatment options such as salicylates, corticosteroids, and immunosuppressants often come with severe side effects, limited bioavailability, development drug resistance, which hampers their therapeutic effectiveness. Therefore, it imperative to explore natural strategies safe alternative treatments for UC. Currently, around 40% UC patients find relief through constituents, can help reduce toxic effects maintain clinical remission. This review aims provide summary both preclinical evidence supporting efficacy various substances prophylaxis These include plant extracts, essential oils, nutraceuticals, phytochemicals. Furthermore, we will delve into potential mechanisms that underlie protective curative actions these novel herbal agents. In summary, this effectiveness remedies UC, shedding light on findings behind actions. alternatives offer hope improved outcomes reduced individuals suffering from challenging condition. Graphical

N-acetylcysteine (NAC), an acetylated derivative of the amino acid L-cysteine, has been widely used as a mucolytic agent and antidote for acetaminophen overdose since 1960s 1980s, respectively. NAC possesses antioxidant, cytoprotective, anti-inflammatory, antimicrobial, properties, making it promising therapeutic wide range diseases in both humans domesticated animals. Oxidative stress inflammation play major role onset progression all these diseases. NAC's primary is to replenish glutathione (GSH) stores, master antioxidant tissues; however, can also reduce levels pro-inflammatory tumor necrosis factor-alpha (TNF-∝) interleukins (IL-6 IL-1β), inhibit formation microbial biofilms destroy biofilms, break down disulfide bonds between mucin molecules. Many experimental studies have conducted on use address pathological conditions; its effectiveness clinical trials remains limited often conflicting results. The purpose this review provide concise overview usages treatment different human domestic animal disorders.

Background/Objectives: Perilla frutescens has historically been used to protect against inflammation and redox stress. This partly attributed its high polyphenolic content; however, components in extract remain incompletely defined. study aimed characterise the composition evaluate effect on transcription factor nuclear erythroid 2-related 2 (Nrf2), regulating antioxidant defenses during oxidative Methods: Hot water extraction from leaves was followed by fractionation using four solvents of different polarity, namely methanol, butanol, ethyl acetate ether. The these fractions analysed RP-HPLC, some compounds were quantified. total phenolic, flavonoid, ortho-diphenolic contents each fraction determined. activity assessed metal cation reduction radical scavenging assays. A dual-luciferase assay a human NQO1 ARE-luciferase reporter plasmid employed quantify Nrf2 activation fractions. Results: HPLC analysis identified 35 compounds, with highest phenolic content present polar rosmarinic acid being major constituent. Radical tests (DPPH ABTS) confirmed capacity On cells vitro, methanol displayed strongest activity, showing up 1.5-fold increase induction. Conclusions: shown that contains diversity contributing potent effects, butanol most efficient solvents. While is expected be contributor towards providing protection stress, further work required synergystic effects between polyphenols.
