Abstract Background Astragaloside IV (AS-IV) is one of the basic components Astragali radix , that has been shown to have preventive effects against various diseases, including cancers. This study aimed explore role AS-IV in hepatocellular carcinoma (HCC) and its underlying mechanism. Methods The cell viability, glucose consumption, lactate production, extracellular acidification rate (ECAR) SNU-182 Huh7 lines were detected by specific commercial kits. Western blot was performed analyze succinylation level lines. interaction between lysine acetyltransferase (KAT) 2 A phosphoglycerate mutase 1 (PGAM1) evaluated co-immunoprecipitation immunofluorescence assays. KAT2A vivo explored using a xenografted tumor model. Results results indicated treatment downregulated protein levels ECAR, decreased AS-IV-treated lines, reversed after overexpression. interacted with PGAM1 promote at K161 site. overexpression promoted viability glycolysis which partly blocked following inhibition. In tumor-bearing mice, suppressed growth though inhibiting KAT2A-mediated PGAM1. Conclusion inhibited HCC regulating PGAM1, suggesting might be potential suitable therapeutic agent for treating HCC.
