Abstract Regarding different medical benefits of fungi, using the mushroom extracts as wound-healing agents is gaining popularity. This study, evaluated wound healing characteristics Trametes versicolor . Anti-oxidant activity addressed by employing DPPH (2,2-diphenyl-1-picrylhydrazyl) assay resulting 53.7% inhibitory effect. Besides, for anti-microbial ability determination, MIC (Minimum Inhibitory Concentration) extract measured which Escherichia coli growth was inhibited at 1.1 mg/ml, and Staphylococcus aureus did not grow 4.38 mg/ml extract. The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) method indicated dose dependence with 63 ± 3% 28 viability 1250 μg/ml 156.25 extract, higher concentration caused cell viability. outcome gene expression analysis determined that overall FGF2 (Fibroblast Growth Factor 2), IL-1β (Interleukin-1β), TGF-β1 (Transforming Factor-β1) 4 times 48 h than 24 in treated cells, suggesting a stimulating effect on growth. An in-vivo animal model suggested enhanced process after treatment 0.01 g Furthermore, number fibroblasts, epidermal thickness, collagen fiber respectively 2, 3, threefold mice when compared to untreated mice. wounds showed 100% 60% within 14 days. results this research show promise fungus-based treatments, may help tissue regeneration cutaneous wounds.

Trametes versicolor is an important fungus with medicinal properties and a significant role in lignocellulose degradation. In this study, we constructed high-quality chromosome-level genome of T. using Illumina, PacBio HiFi, Hi-C sequencing technologies. The assembled 47.42 Mb size contains 13,307 protein-coding genes. BUSCO analysis revealed gene completeness results 95.80% 95.90%, respectively. Phylogenetic showed that most closely related to pubescens, followed by cinnabarina coccinea. Comparative genomic identified 266 syntenic blocks between Wolfiporia cocos, indicating conserved evolutionary pattern the two species. Gene family highlighted expansion contraction genes functional categories biosynthesis secondary metabolites, including several versicolor-specific Key involved degradation triterpene production were within CAZyme CYP450 families. Transcriptomic under dark light conditions changes expression metabolism, suggesting signals regulate metabolic pathways. A total 2577 transporter proteins 2582 membrane mapped genome, 33 metabolite clusters identified, light-sensitive clusters. This study offers comprehensive resource for further investigation into genomics, regulation, versicolor, providing valuable insights fungal evolution biotechnological applications.

Coriolus versicolor (CV), known in traditional Chinese medicine for over 2000 years, is currently used China and Japan to reduce chemotherapy or radiotherapy side effects cancer patients. Despite extensive research, its still need improvement. This study aimed determine if combining CV extract with LY294002, an inhibitor of the phosphatidylinositol-3-kinase (PI3K) signalling pathway, enhances cell treatment, potentially leading a novel therapeutic approach. Three human lines (MCF-7, HeLa, A549) were treated alone combined LY294002. Cell viability was assessed using MTT assays. Then, HeLa MCF-7 cells most sensitive co-treatment evaluate colony formation, apoptosis, cycle, migration invasion, phospho-PI3K expression. The results demonstrated that LY294002 enhanced extract's anti-tumour by reducing formation. treatment more effectively induced G0/G1 cycle arrest, promoted reduced invasion migration, inhibited expression compared each agent alone. highlights potent cytotoxic enhancement between on cells, primarily inhibiting These findings suggest promising avenues developing combination therapies targeting cancer.
