High-sensitivity and high-specificity biodetection is critical for advancing applications in life sciences, biosafety, food safety, environmental monitoring. CRISPR/Cas systems have emerged as transformative tools biosensing due to their unparalleled specificity, programmability, unique enzymatic activities. They exhibit two key cleavage behaviors: precise ON-target guided by specific protospacers, which ensures accurate target recognition, bystander activity triggered upon binding, enables robust signal amplification. These properties make highly versatile designing biosensors ultra-sensitive detection. This review comprehensively explores recent advancements system-based biosensors, highlighting impact on improving performance. We discuss the integration of with diverse readout mechanisms, including electrochemical, fluorescent, colorimetric, surface-enhanced Raman scattering (SERS), so on. Additionally, we examine development integrated systems, such microfluidic devices portable leverage technology point-of-care testing (POCT) high-throughput analysis. Furthermore, identify unresolved challenges, aiming inspire innovative solutions accelerate translation these technologies into practical diagnostics, food, environment safety.

Protein function investigation and clinical assay are fundamental to modern biology medical diagnostics. Flap endonuclease 1 (FEN1), a key enzyme in DNA replication repair, plays critical role the progression of many diseases. Taking FEN1 as an example, we present novel protein detection platform combining triboelectric nanogenerator (TENG) CRISPR/Cas technologies. As specific form TENG, transistor-droplet electricity generator (TDEG) is explored, which offers low-cost, simple fabrication approach with real-time capability. Meanwhile, activated system catalyzes reactions on three-dimensional tetrahedron interface, promising high sensitivity. This work not only demonstrates powerful method for rapid but also pioneers integration TENG. It has great prospect future development TENG sensors.

Abstract Organic photoelectrochemical transistor (OPECT) biosensors have emerged as prevalent candidates for bioanalytical technologies because of their intrinsic amplification properties and excellent detection performance. Herein, a novel OPECT biosensor analyzing the RNA demethylation protein ALKBH3 is developed. A BiOI/In 2 O 3 type‐II heterojunction employed photoactive material gate, an organic semiconductor, poly(3,4‐ethylenedioxythiophene): poly(styrenesulfonic) (PEDOT:PSS), used channel modification material. ALKBH3‐mediated demethylation, switch, triggered trans‐cleavage activity CRISPR‐Cas12a system, which subsequently leads to failure in formation bipedal DNA walker. Contrarily, ferrocenecarboxylic acid (FcA), acts effective electron donor, cannot be captured, leading modulation gating characteristics degradation current response. Compared conventional method, exhibits higher sensitivity lower limit protein, thereby facilitating monitoring changes under effect environmental pollutants demonstrating crucial biomarker potential evaluating its toxicological effects.

CRISPR technology has revolutionized genome editing by enabling precise, permanent modifications to genetic material. To circumvent the irreversible alterations associated with traditional methods and facilitate research on both essential nonessential genes, interference or inhibition (CRISPRi) activation (CRISPRa) were developed. The gene-silencing approach leverages an inactivated Cas effector protein paired guide RNA obstruct transcription initiation elongation, while gene-activation exploits programmability of activate gene expression. Recent advances in CRISPRi technology, combination other technologies (e.g., biosensing, sequencing), have significantly expanded its applications, allowing for genome-wide high-throughput screening (HTS) identify determinants phenotypes.
