Physiological and psychological stressors can exert wide-ranging effects on the human brain behavior. Research has improved understanding of how sympatho-adreno-medullary (SAM) hypothalamic-pituitary-adrenocortical (HPA) axes respond to differential responses that occur depending stressor type. Although physiological function SAM HPA is promote survival safety, exaggerated psychobiological reactivity in psychiatric disorders. Exaggerated may more for certain types stressors, specifically, psychosocial stressors. Understanding body regulates these provide insight into ways be modulated. Non-invasive neuromodulation one way responding altered; research interventions further insights circuits modulate stress reactivity. This review focuses acute might effective altering considerable treating pathology, it imperative first understand mechanisms non-clinical populations; therefore, this will emphasize populations with no known pathology consider results translate those pathologies.

Late-life depression (LLD) is one of the most common mental disorders among older adults. Population aging, social stress, and COVID-19 pandemic have significantly affected emotional health adults, resulting in a worldwide prevalence LLD. The clinical phenotypes between LLD adult differ terms symptoms, comorbid physical diseases, coexisting cognitive impairments. Many pathological factors such as imbalance neurotransmitters, decrease neurotrophic factors, an increase β-amyloid production, dysregulation hypothalamic-pituitary-adrenal axis, changes gut microbiota, are allegedly associated with onset However, exact pathogenic mechanism underlying remains unclear. Traditional selective serotonin reuptake inhibitor therapy results poor responsiveness side effects during treatment. Neuromodulation therapies complementary integrative been proven safe effective for treatment Importantly, pandemic, modern digital intervention technologies, including socially assistive robots app-based interventions, to be advantageous providing personal services patients

Abstract Deep brain stimulation (DBS) modulates local and widespread connectivity in dysfunctional networks. Positive results are observed several patient populations; however, the precise mechanisms underlying treatment remain unknown. Translational DBS studies aim to answer these questions provide knowledge for advancing field. Here, we systematically review literature on involving models of neurological, developmental neuropsychiatric disorders a synthesis current scientific landscape surrounding this topic. A systematic analysis was performed following PRISMA guidelines. 407 original articles were included. Data extraction focused study characteristics, including protocol, behavioural outcomes, action. The number published increased over years, 16 rat 13 mouse transgenic or healthy animals exposed external factors induce symptoms. Most targeted telencephalic structures with varying settings. outcomes reported 85.8% included studies. In psychiatric neurodevelopmental disorders, DBS-induced effects associated changes monoamines neuronal activity along mesocorticolimbic circuit. For movement improves symptoms via modulation striatal dopaminergic system. dementia epilepsy models, cellular molecular aspects hippocampus shown underlie symptom improvement. Despite limitations translating findings from preclinical clinical settings, rodent have contributed substantially our pathophysiology disease mechanisms. Direct inhibition/excitation neural activity, whereby pathological oscillatory within networks, is among major theories its mechanism. However, there fundamental mechanisms, optimal targets parameters that need be better understood improve therapy more individualized according patient’s predominant

Background In randomized clinical trials (RCTs) investigating the application of transcranial alternating current stimulation (tACS) in schizophrenia, inconsistent results have been reported. The purpose this exploratory systematic review RCTs was to evaluate tACS as an adjunct treatment for patients with schizophrenia based on its therapeutic effects, tolerability, and safety. Methods Our analysis included that evaluated adjunctive tACS’ effectiveness, safety patients. Three independent authors extracted data synthesized it using RevMan 5.3 software. Results involving 76 were encompassed analysis, 40 participants receiving active 36 sham tACS. study revealed a significant superiority over improving total psychopathology (standardized mean difference [SMD] = −0.61, 95% confidence interval [CI]: −1.12, −0.10; I 2 16%, p 0.02) negative (SMD −0.65, CI: −1.11, −0.18; 0%, 0.007) schizophrenia. two groups, however, showed no differences positive psychopathology, general or auditory hallucinations (all > 0.05). Two examined neurocognitive effects tACS, yielding varied findings. Both groups demonstrated similar rates discontinuation due any reason adverse events Conclusion Adjunctive is promising viable approach mitigating individuals diagnosed However, gain more comprehensive understanding tACS’s imperative conduct extensive, meticulously planned, well-documented RCTs.

Abstract While neurotechnology provides opportunities for novel mental health interventions, preferences of patients and the general public, reasons their choices are still unclear. Here, we conducted a large-scale online survey with 784 participants, half them suffering from psychiatric and/or neurological conditions. We asked about techniques ranging invasive (pharmaceutical drugs brain implants) to noninvasive approaches (ultrasound, magnetic, or electric stimulation). First, participants had low level prior knowledge but were interested excited these opportunities. Second, both population preferred focused ultrasound stimulation (first choice) while implants ranked 3 rd 5 th out five, respectively. Finally, that preference was mainly driven by whether they perceived technology as safe, rather than effective. Overall, this identifies safety main criterion interventions shows such neuromodulation. Highlights Focused is intervention conditions The consistent between healthy Getting more information different neuromodulation reduced confusion increased feelings being excited, optimistic, comfortable ranking linked less related efficiency indicating important

The habenula (Hb) is a small, evolutionarily conserved epithalamic structure implicated in functions such as reward and mood regulation. Prior imaging work suggests that Hb's structural functional properties may relate to treatment response depression other disorders. We used multimodal MRI techniques investigate the potential involvement of Hb subcallosal cingulate area deep brain stimulation (SCC-DBS) for treatment-resistant Using an automated segmentation technique, we compared volume at baseline subsequent post-operative timepoint (4.4 ± 3.0 years after surgery) cohort 32 patients who received SCC-DBS. Clinical (≥50% decrease HAMD-17 from 12 months post-operation) was significantly associated with longitudinal change: responders tended have increased over time, while non-responders showed decreased (t = 2.4, p 0.021). additionally (fMRI) subcohort SCC-DBS (n 12) immediate within-patient changes connectivity stimulation. Active DBS several prefrontal corticolimbic regions (TFCE-adjusted pBonferroni < 0.0001), many which been previously neurocircuitry depression. Taken together, our results suggest play important role antidepressant effect

Abstract Various medication options with good efficacy are currently available for bipolar disorder treatment, but there some major pitfalls regarding these agents. Therefore, improving the treatment outcome in is a pressing issue, and it involves both optimizing interventions pursuing novel options. Multiple pharmacological nonpharmacological therapies acting through different mechanisms have been investigated as potential therapeutic targets disorder. Studies on of treatments shown promising results most challenging areas disorder, including depressive episodes cognitive functional impairment. Nevertheless, them still need to be tested larger samples rigorously designed randomized controlled trials before they can confidently transferred clinical practice. Future studies should follow enriched protocols further stratification allow homogeneous groups modalities.
