Photoreceptor cell death and inflammation are known to occur progressively in retinal degenerative diseases, however the molecular mechanisms underlying these biological processes largely unknown. Extracellular vesicles (EV) essential mediators of cell-to-cell communication with emerging roles modulation immune responses. EVs including exosomes encapsulate transfer nucleic acids, microRNA (miRNA), recipient cells which disease may result dysfunctional responses a loss homeostatic regulation. In this work we investigated role isolated small-medium sized EV (s-mEV) includes both healthy degenerating retina. Isolated s-mEV from normal retinas were characterized using dynamic light scattering, transmission electron microscopy western blotting, quantified across 5 days photo-oxidative damage-induced degeneration nanotracking analysis. Small RNAseq was used characterize miRNA cargo damaged retinas. Finally, effect exosome inhibition on conducted systemic daily administration inhibitor GW4869 situ hybridization s-mEV-abundant miRNA, miR-124-3p. Electroretinography immunohistochemistry performed assess functional morphological changes retina as GW4869-induced depletion. Results demonstrated an inverse correlation between secretion photoreceptor survivability, decrease numbers following degeneration. revealed that s-mEVs contained uniquely enriched miRNAs comparison whole tissue however, there no differential change miRNAnome damage. Exosome via use also found exacerbate degeneration, reduced function increased levels damage exosome-inhibited mice. Further, GW4869-treated mice displayed impaired translocation photoreceptor-derived miR-124-3p inner during Taken together, propose their play maintaining homeostasis through immune-modulation, have potential be targeted gene therapy for diseases.

Autoimmune cytopenias (AICy) and autoimmune diseases (AID) can complicate both lymphoid myeloid neoplasms, often represent a diagnostic therapeutic challenge. While hemolytic anemia (AIHA) immune thrombocytopenia (ITP) are well known, other rarer AICy (autoimmune neutropenia, aplastic anemia, pure red cell aplasia) AID (systemic lupus erythematosus, rheumatoid arthritis, vasculitis, thyroiditis, others) poorly recognized. This review analyses the available literature of last 30 years regarding occurrence AICy/AID in different onco-hematologic conditions. The latter include chronic lymphocytic leukemia (CLL), lymphomas, multiple myeloma, myelodysplastic syndromes (MDS), myelomonocytic (CMML), myeloproliferative acute leukemias. On whole, observed up to 10% CLL with higher frequencies certain subtypes non-Hodgkin lymphoma, whilst they occur less than 1% low-risk MDS CMML. described 30% patients, including immune-mediated hemostatic disorders (acquired hemophilia, thrombotic thrombocytopenic purpura, anti-phospholipid syndrome) that may be severe fatal. Additionally, found about patients receiving hematopoietic stem transplant or treatment new checkpoint inhibitors. Besides difficulties, these clinical management already immunocompromised patients.

Systemic lupus erythematosus (SLE) is a common autoimmune connective tissue disease with unclear etiology and pathogenesis. Mesenchymal stem cell (MSC) MSC derived extracellular vesicles (EVs) play important roles in regulating innate adaptive immunity, which are involved many physiological pathological processes contribute to the immune homeostasis SLE. The effects of MSCs EVs on SLE have been drawing more attention during past few years. This article reviews immunomodulatory underlying mechanisms MSC/MSC-EVs SLE, provides novel insight into understanding pathogenesis guiding biological therapy.

Abstract: Systemic sclerosis (SSc) is a complex autoimmune disease characterized by vascular damage, vasoinstability, and decreased perfusion with ischemia, inflammation, exuberant fibrosis of the skin internal organs. Biomarkers are analytic indicators biological processes within an individual that can be accurately reproducibly measured. The field biomarkers in SSc as recent studies have implicated at least 240 pathways dysregulated proteins pathogenesis. Anti-nuclear antibodies (ANA) classical well-described clinical classifications present more than 90% patients include anti-centromere, anti-Th/To, anti-RNA polymerase III, anti-topoisomerase I antibodies. Transforming growth factor-β (TGF-β) central to fibrotic process intimately intertwined other biomarkers. Tyrosine kinases, interferon-1 signaling, IL-6 endogenous thrombin, peroxisome proliferator-activated receptors (PPARs), lysophosphatidic acid receptors, amino metabolites new potential for developing therapeutic agents. The etiopathogenesis of autoimmune skin diseases is complex and still not fully understood. role epigenetic factors emphasized in the development such diseases. MicroRNAs (miRNAs), a group non-coding RNAs (ncRNAs—non-coding RNAs), are one important post-transcriptional factors. miRNAs have significant regulation immune response by participating process differentiation activation B T lymphocytes, macrophages, dendritic cells. Recent advances research on provided new insights into pathogenesis potential diagnostic therapeutic targets many pathologies. Numerous studies revealed change expression some microRNAs inflammatory disorders, miRNA promising goal. Systemic lupus erythematosus (SLE) is a systemic autoimmune disease characterized by inflammation and abnormal production of autoantibody, but the mechanisms aberrant immune responses are currently unknown. Recently, growing evidence has suggested that infection plays pivotal role in SLE. Here, we investigate infectious agents (e.g., Epstein-Barr virus, parvovirus B19, human T-lymphotropic virus type 1, immunodeficiency endogenous retroviruses) pathogenesis More importantly, explore known underlying involvement SLE, including molecular mimicry, epitope spreading, superantigen production, bystander activation, persistent viral infection, altered apoptosis, clearance deficiency, epigenetic alterations DNA methylation microRNAs). However, some malaria parasites, hepatitis B Toxoplasma gondii, Helicobacter pylori) may exert protective effects on Therefore, relationship between SLE multifaceted multidirectional, causative and/or associations, which warrant further investigation future.

Many studies indicate that microRNAs (miRNAs) could be potential biomarkers for various diseases. The purpose of this study was to investigate the clinical value serum exosomal miRNAs in systemic lupus erythematosus (SLE).Serum exosomes were isolated from 38 patients with SLE and 18 healthy controls (HCs). expression miR-21, miR-146a miR-155 within examined by reverse transcription-quantitative polymerase chain reaction (RT-qPCR). Using receiver operating characteristic (ROC) curves, we evaluated diagnostic miRNAs.Exosomal miR-21 upregulated (p<0.01), whereas (p<0.05) downregulated SLE, compared HCs. (p<0.01) higher nephritis (LN) than those without LN (non-LN). analysis ROC curves revealed showed a LN. Furthermore, (R=0.44, p<0.05) (R=0.33, positively correlated proteinuria. negatively associated anti-SSA/Ro antibodies (R=-0.38, p<0.05), anti-dsDNA (R=-0.39, p<0.05).These findings suggested levels may serve as diagnosis
