Abstract Background Impaired osteo-/angiogenesis, excessive inflammation, and imbalance of the osteoimmune homeostasis are involved in pathogenesis alveolar bone defect caused by periodontitis. Unfortunately, there is still a lack ideal therapeutic strategies for periodontitis that can regenerate while remodeling microenvironment. Quercetin, as monomeric flavonoid, has multiple pharmacological activities, such pro-regenerative, anti-inflammatory, immunomodulatory effects. Despite its vast spectrum quercetin’s clinical application limited due to poor water solubility low bioavailability. Results In this study, we fabricated quercetin-loaded mesoporous bioactive glass (Quercetin/MBG) nano-delivery system with function continuously releasing quercetin, which could better promote regeneration regulate immune microenvironment compared pure MBG treatment. particular, effectively decreased injection frequency quercetin yielding favorable results. view above excellent effects achieved sustained release further investigated mechanisms. Our findings indicated under microenvironment, intervention restore osteo-/angiogenic capacity periodontal ligament stem cells (PDLSCs), induce regulation macrophages exert an osteoimmunomodulatory effect. Furthermore, also found via be partially blocked overexpression key microRNA——miR-21a-5p, worked through inhibiting expression PDCD4 activating NF-κB signaling pathway. Conclusion summary, our study shows potential approach reconstructing defects it offers new perspective treating miR-21a-5p thereby creating Graphical

The intestine is essential for the modulation of nutrient absorption and removal waste. Gut pathologies, such as cancer, inflammatory bowel diseases (IBD), irritable syndrome (IBS), celiac disease, which extensively impact gut functions, are thus critical human health. Targeted drug delivery to tackle these diseases, improve therapy efficacy, minimize side effects. Recent strategies have taken advantage both active passive nanocarriers, designed protect until it reaches correct site modulate release via use different physical–chemical strategies. In this systematic review, we present a literature overview nanocarriers used in set chronic intestinal highlighting rationale behind controlled therapies. overall aim provide reader with useful information on current approaches targeting novel therapeutic

The intestinal barrier maintained by various types of columnar epithelial cells, plays a crucial role in regulating the interactions between contents (such as microbiota), immune system, and other components. Dysfunction mucosa is significant pathophysiological mechanism clinical manifestation inflammatory bowel disease (IBD). However, current therapies for IBD primarily focus on suppressing inflammation, no disease-modifying treatments specifically target barrier. Given side effects associated with chronic immunotherapy, effective alternative that promote mucosal healing are highly attractive. In this review, we examined function mechanisms behind its disruption IBD. We illustrated complex process proposed therapeutic approaches to strategies These included application stem cell transplantation organ-like tissue engineering generate new tissue. Finally, discussed potential restore treatment

Background Respiratory allergy is a serious respiratory disorder characterized by inflammation, mucus hypersecretion, and airway tissue sclerosis. Disruption of the T helper 1 (Th1) 2 (Th2) immune systems stimuli induced house dust mites (HDM) fine particulate matter leads to secretion various inflammatory cytokines, resulting in diseases inflammation. Chitooligosaccharides (COS) are known for their antioxidant anti-inflammatory properties. Methods Human epithelial cells (BEAS-2B) were cultured DMEM/F12 medium containing COS at concentrations 25–100 µg/ml 24 h. No intracellular toxicity was observed up 1,000 µg/ml. Cell experiments conducted below 100 µg/ml, while animal performed mg/kg body weight 4 weeks. Samples right lung obtained from experimental animals used gene protein expression analysis, whereas samples contralateral immunohistochemical analysis. Results regulated Th1 immunity inhibiting major including tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6), BEAS-2B cells. In HDM-induced allergic model, suppressed infiltration around airways inhibited mRNA cytokines tissues, also reducing nuclear factor kappa B (NF-κB)-related proteins. Furthermore, results confirmed suppression levels immunoglobulin E (IgE) blood secreted mast activated HDM, which led reduction hypersecretion Conclusion summary, thought improve resistance alleviating caused HDM regarded as substances that regulate balance Th2 affected hypersecretion.

Chemotherapy is widely recognized as a highly efficacious modality for cancer treatment, involving the administration of chemotherapeutic agents to target and eradicate tumor cells. Currently, oral stands prevailing utilized method delivering chemotherapy drugs. However, majority anti-tumor medications exhibit limited solubility permeability, poor stability in harsh gastrointestinal environments, thereby impeding their therapeutic efficacy chemotherapy. Therefore, more micro-nano drug delivery carriers have been developed used effectively deliver anti-cancer drugs, which can overcome physiological barriers, facilitate administration, ultimately improve efficacy. In this paper, we first discuss effects various biological barriers on approach. Then, development based biomedical components, such micelles, dendrimers, hydrogels, liposomes, inorganic nanoparticles, etc. were introduced. Finally, current dilemma potential clinical treatment discussed. The primary objective review introduce methods serve point reference advancement novel carriers, with ultimate goal informing future applications.
