In this present study, we developed and characterized a series of supramolecular G4 hydrogels by integrating β-cyclodextrin (β-CD) boronic acid linkers into matrix to enhance antibacterial activity against Staphylococcus aureus (S. aureus). We systematically investigated how varying the number free moieties (ranging from two six), along with guanosine β-CD content, influences both structural integrity antimicrobial efficacy these materials. Comprehensive characterization using FTIR, circular dichroism, X-ray diffraction, SEM, AFM, rheological measurements confirmed successful synthesis revealed that higher content correlated stronger, more organized network. The most effective hydrogel displayed an inhibition zone 25 mm in disk diffusion assays, was further explored as drug delivery platform, aim exploit capacity cavity incorporate hydrophobic drugs. Norfloxacin (Nfx), poorly water-soluble antibiotic, successfully encapsulated within through inclusion complex formation β-CD, improving its solubility enabling sustained, targeted release. Nfx-loaded expanded 49 completely eradicated S. cells 24 h, outperforming unloaded Nfx. These results highlight synergistic effect controlled release, underlining potential versatile platforms for localized therapy, such wound dressings or implant coatings. Nevertheless, vivo studies long-term stability assessments are needed fully establish clinical relevance, safety, scalability before systems can be translated routine healthcare applications.
