ABSTRACT In spite of great advances in modern medicine, there are a few effective strategies for the treatment neurodegenerative diseases characterised by neuron loss or degeneration. This results from complex pathogenesis and limited drug uptake brain due to presence blood‐brain barrier. Nanoparticle‐based delivery systems expected improve utilisation. Nucleic acid aptamers, known as chemical antibodies, demonstrate remarkable affinity and specificity for targets. Therefore, aptamers are proposed an alternative to antibody in extensive applications. However, nucleic exhibit poor tolerance degradation by nucleases, which severely hampers their biological Herein, we developed a regulation pattern utilizing small-molecule-mediated terminal manipulation, could prevent the interaction of DNA with exonucleases help persist desired conformation high stability. Diagonal T-T bases were designed ends be chemically cross-linked trioxsalen via photocatalyzed cycloaddition. Aptamers different patterns cross-linking sites synthesized. Experimental investigation molecular dynamics simulations combinedly revealed that efficiency depended on multiple factors: (i) number sequences, (ii) spatial (iii) competitive binding ability compared other base sites. The locked exhibited superior exonuclease resistance, especially both 3'- 5'-cross-linked ends, thus demonstrating great target capability. Notably, application exploration, bound receptor-binding domains SARS-CoV-2, showed performance virus infection inhibition.
