Abstract Nanomedicines are extensively used in cancer therapy. Covalent organic frameworks (COFs) crystalline porous materials with several benefits for therapy, including porosity, design flexibility, functionalizability, and biocompatibility. This review examines the use of COFs therapy from perspective reticular chemistry function‐oriented design. First, modification sites functionalization methods discussed, followed by their potential as multifunctional nanoplatforms tumor targeting, imaging, integrating functional components. Finally, some challenges clinical translation presented hope promoting development COF‐based anticancer nanomedicines bringing closer to trials.

Detection of circulating tumor DNA (ctDNA) in liquid biopsy is great importance for diagnosis but difficult due to its low amount bodily fluids. Herein, a novel ctDNA detection platform established by quantifying amplification by-product pyrophosphate (PPi) using newly designed bivariable lanthanide metal–organic framework (Ln-MOF), namely, Ce/Eu-DPA MOF (CE-24, DPA = pyridine-2,6-dicarboxylic acid). CE-24 exhibits ultrafast dual-response (fluorescence enhancement and enzyme-activity inhibition) PPi stimuli virtue host–guest interaction. The applied detecting colon carcinoma-related (KARS G12D mutation) combined with the isothermal nucleic acid exponential reaction (EXPAR). triggers generation large PPi, quantification achieved through ratio fluorescence/colorimetric dual-mode assay PPi. combination EXPAR sensing allows method be low-cost, convenient, bioreaction-compatible (freedom from interference bioreaction systems), sensitive (limit down 101 fM), suitable on-site detection. To best our knowledge, this work first application Ln-MOF detection, it provides universal strategy rapid biomarkers point-of-care scenarios.
