Accurate quantitative detection of DNA is an advanced strategy in various fields (such as disease diagnosis and environmental monitoring), but the classical method usually suffers from low sensitivity, expensive thermal cyclers, or strict annealing conditions. Herein, a MOF-ERA platform for ultrasensitive HBV-DNA constructed by integrating metal-organic framework (MOF)-mediated double energy transfer nanoprobe with exonuclease III (Exo III)-assisted target recycling amplification. The proposed containing donor two receptors simply composed MOFs (UiO-66-NH2, well-studied MOF) modified signal probe formed hybridization carboxyuorescein (FAM)-labeled (FDNA) black hole quencher (BHQ1)-terminated (QDNA), resulting fluorescence signal. After addition HBV-DNA, Exo degradation to FDNA activated, leading liberation numerous FAM molecules, followed generation significant owing negligible binding free molecules. results certify that can be successfully used assay range 1.0-25.0 nM limit 97.2 pM, which lower than without BHQ1 III. superiorities background high selectivity holds promise early clinical biomedicine applications.
