Abstract Background Wastewater treatment plants contribute approximately 6% of anthropogenic methane emissions. Methanotrophs, capable converting into polyhydroxybutyrate (PHB), offer a promising solution for utilizing as carbon source, using activated sludge seed culture PHB production. However, maintaining and enriching PHB-accumulating methanotrophic communities poses challenges. Results This study investigated the potential Methylosinus trichosporium OB3b to bioaugment consortium within enhance Waste-activated sludges with varying ratios M. (1:0, 1:1, 1:4, 0:1) were cultivated. The results revealed substantial growth consumption in waste-activated OB3b-amended cultures, particularly 1:1 ratio. Enhanced accumulation, reaching 37.1% same ratio culture, indicates dominance Type II methanotrophs. Quantification methanotrophs by digital polymerase chain reaction showed gradual increases methanotrophs, correlating increased while initial bioaugmentation was observed, its presence decreased subsequent cycles, indicating other Microbial community analysis highlighted successful enrichment methanotrophs-dominated cultures due addition OB3b, outcompeting I Methylocystis Methylophilus spp. most abundant cultures. Conclusions Bioaugmentation strategies, leveraging could significantly production foster sludge. These findings integrating wastewater plants, providing sustainable resource recovery.
