ABSTRACT The ability to observe the social behavior of others and use observed information bias future action is a fundamental building block cognition 1,2 . A foundational question whether observation experience engage common circuit mechanisms that enable behavioral change. While classic studies on learning have shown aggressive behaviors can be learned through 3 , it remains unclear aggression promotes persistent neural changes generalize new contexts. Here, directly compare effects at brain-wide scale, we develop strategy perform large-scale cell-type specific recordings across subcortical networks for control learning. We record longitudinally while animals “train” direct or observation, then probe shared differences in activity novel “hard” context. Using supervised unsupervised methods quantification, detect unique signatures not present with no training. During find widespread activation mimics generation, critical signals associated reward threat After persist into context, increased similarity dynamics between groups. Network-level modeling reveals core network, decoupling inhibition from key hypothalamic output region. This demonstrates “experience-like” during recruit plasticity mechanism biases toward adaptive defensive strategies

Fiber photometry is a key technique for characterizing brain-behavior relationships in vivo. Initially, it was primarily used to report calcium dynamics as proxy neural activity via genetically encoded indicators. This generated new insights into brain functions including movement, memory, and motivation at the level of defined circuits cell types. Recently, opportunity discovery with fiber has exploded development an extensive range fluorescent sensors biomolecules neuromodulators peptides that were previously inaccessible critical advance, combined availability affordable "plug-and-play" recording systems, made monitoring molecules high spatiotemporal precision during behavior highly accessible. However, while opening exciting avenues research, rapid expansion applications occurred without coordination or consensus on best practices. Here, we provide comprehensive guide help end-users execute, analyze, suitably interpret studies.

Sexual, parental, and aggressive behaviors are central to the reproductive success of individuals species survival thus supported by hardwired neural circuits. The behavior control column (RBCC), which comprises medial preoptic nucleus (MPN), ventrolateral part ventromedial hypothalamus (VMHvl), ventral premammillary (PMv), is essential for all social behaviors. RBCC integrates diverse hormonal metabolic cues adjusts an animal's physical activity, hence chance encounters. further engages mesolimbic dopamine system maintain interest reinforces actions that time-locked with We propose brainstem form a dual-control generating moment-to-moment actions. Male sexual behavior is innate and rewarding. Despite its centrality to reproduction, a molecularly specified neural circuit governing male reward remains be characterized. We have discovered developmentally wired necessary sufficient for mating. This connects chemosensory input BNSTprTac1 neurons, which innervate POATacr1 neurons that project centers regulating motor output reward. Epistasis studies demonstrate are upstream of BNSTprTac1-released substance P following mate recognition potentiates activation through Tacr1 initiate Experimental triggers mating, even in sexually satiated males, it rewarding, eliciting dopamine release self-stimulation these cells. Together, we uncovered governs the key aspects behavior: displays, drive,

The hypothalamus plays a crucial role in regulating sleep-wake cycle and motivated behavior. Sleep disturbance is associated with impairment cognitive affective functions. However, how hypothalamic dysfunction may contribute to inter-related sleep, cognitive, emotional deficits remain unclear. We curated the Human Connectome Project dataset investigated resting state functional connectivities (rsFC) were sleep dysfunction, as evaluated by Pittsburgh Quality Index (PSQI), performance, subjective mood states 687 young adults (342 women). Imaging data processed published routines corrected threshold. We examined inter-relationship amongst rsFC, PSQI score, clinical measures mediation analyses. In whole-brain regressions age drinking severity covariates, men showed higher rsFC right insula correlation score. No clusters identified women at same Both hypothalamic-insula score significantly correlated anxiety depression scores men. Further, analyses that mediated relationship between anxiety/depression symptom bidirectionally men.Sleep negative emotions insula, core structure of interoceptive circuits. Notably, anxiety-depression altered are related poor quality. These findings specific men, suggesting potential sex differences neural circuits need be further investigated.
