Ultrasound is highly biopermeable and can non-invasively penetrate deep into the brain. Stimulation with patterned low-intensity ultrasound induce sustained inhibition of neural activity in humans animals, potential implications for research therapeutics. Although mechanosensitive channels are involved, cellular molecular mechanisms underlying neuromodulation by remain unknown. To investigate mechanism action stimulation, we studied effects two types on synaptic transmission network using whole-cell recordings primary cultured hippocampal cells. Single-shot pulsed-wave (PW) or continuous-wave (CW) had no effect activity. By contrast, although repetitive CW stimulation also effect, PW persistently reduced spontaneous recurrent burst firing. This inhibitory was dependent extrasynaptic—but not synaptic—GABA A receptors, abolished under astrocyte-free conditions. Pharmacological activation astrocytic TRPA1 mimicked increasing tonic GABA current induced ambient GABA. blockade ultrasound. These findings suggest that used our study does have a direct function but instead exerts its neuromodulatory through modulation levels via characteristics TRPA1, which expressed astrocytes.

Low-frequency transcranial ultrasound stimulation (TUS) allows to alter brain functioning with a high spatial resolution and reach deep targets. However, the time-course of TUS effects remains largely unknown. We applied on three targets for different monkeys: anterior medial prefrontal cortex, supplementary motor area perigenual cingulate cortex.
