Fresh fruits and vegetables are potential reservoirs for antimicrobial resistance determinants, but few studies have focused specifically on organic vegetables. The present study aimed to determine the presence of third-generation cephalosporin (3GC)- carbapenem-resistant Gram-negative bacteria fresh produced in city Valencia (Spain). Main expanded spectrum beta-lactamase (ESBL)- carbapenemase-encoding genes were also detected isolates. One hundred fifteen samples analyzed using selective media supplemented with cefotaxime meropenem. Resistance assays twelve relevant antibiotics medical use performed a disc diffusion test. A total 161 isolates tested. Overall, 33.5% presented multidrug 16.8% resistant all β-lactam Imipenem was observed 18% isolates, low levels found ceftazidime Opportunistic pathogens such as Acinetobacter baumannii, Enterobacter spp., Raoultella sp., Stenotrophomonas maltophilia detected, presenting high rates resistance. PCR revealed blaVIM be most frequently isolated ESBL-encoding gene, followed by blaTEM blaOXA-48. These results confirm act 3GC- carbapenem-producing ARB. Further must carried out impact raw food spread AMRs into community.

Abstract Mining is among the human activities with widest environmental impacts, and mining-impacted environments are characterized by high levels of metals that can co-select for antibiotic resistance genes (ARGs) in microorganisms. However, ARGs still poorly understood. Here, we conducted a comprehensive study such worldwide, taking advantage 272 metagenomes generated from global-scale data collection two national sampling efforts China. The average total abundance globally distributed studied mine sites was 1572 times per gigabase, being rivaling urban sewage but much higher than freshwater sediments. Multidrug accounted 40% ARG abundance, tended to co-occur multimetal genes, were highly mobile (e.g. on 16% occurring plasmids). Among 1848 high-quality metagenome-assembled genomes (MAGs), 85% carried at least one multidrug gene plus gene. These ARG-carrying MAGs considerably expanded phylogenetic diversity hosts, providing first representatives Archaea domain three bacterial phyla. Moreover, 54 identified as potential pathogens. Our findings suggest worldwide underexplored hotspots genes.

Soil is recognized as the major reservoir of antibiotic resistance genes (ARGs), harboring most diverse naturally evolved ARGs on planet. Multidrug are a class ARGs, and their high prevalence in natural soil ecosystems has recently raised concerns. Since these express proton motive force (PMF) driven efflux pumps, studying whether pH determinant for selection multidrug pump thus shaping resistome great interest. In this study, we collected 108 soils with values ranging from 4.37 to 9.69 multiple profiled composition metagenomes metagenome-assembled genomes. We observed enriched acidic resistome, abundances have significant dependence. This reflects benefits activity genes, especially PMF-driven inner membrane transferase. addition, preliminary indicate putative microbial participants by applying ecological analyzing tools such stepwise regression random forest model fitting. The decisive influence more impactful than any other examined factors, consequence, revisited edaphic factors resistome; i.e., deterministic mechanisms could lead bottom-up ARG composition. Such developing herein suggested be considered assessing human-driven transmissions.

Abstract Antibiotic resistance genes (ARGs) and metal(loid) (MRGs) coexist in organic fertilized agroecosystems based on their correlations abundance, yet evidence for the genetic linkage of ARG-MRGs co-selected by fertilization remains elusive. Here, an analysis 511 global agricultural soil metagenomes reveals that correlates with a threefold increase number diverse types ARG-MRG-carrying contigs (AMCCs) microbiome (63 types) compared to non-organic soils (22 types). Metatranscriptomic data indicates increased expression AMCCs under higher arsenic stress, co-regulation ARG-MRG pairs. Organic heightens coexistence genomic elements through impacting properties ARG MRG abundances. Accordingly, comprehensive map was constructed delineate distribution coexistent virulence factors mobile metagenome-assembled genomes from lands. The unveils heightened relative abundance potential pathogenicity risks (range 4-6) spread Central North America, Eastern Europe, Western Asia, Northeast China other regions, which acquire risk range 1-3. Our findings highlight co-selects genetically linked ARGs MRGs microbiome, underscore need mitigate these co-resistant safeguard public health.

Abstract Antimicrobial resistance (AMR) poses a severe threat to global health. The wide distribution of environmental antibiotic genes (ARGs), which can be transferred between microbiota, especially clinical pathogens and human commensals, contributed significantly AMR. However, few databases on the spatiotemporal distribution, abundance, health risk ARGs from multiple environments have been developed, absolute level. In this study, we compiled ARG occurrence data generated by high-throughput quantitative PCR platform 1,403 samples in 653 sampling sites across 18 provinces China. database possessed 291,870 records five types habitats abundance 290 ARGs, as well 8,057 30 mobile genetic elements (MGEs) 2013 2020. The global prevalence of antibiotic resistance genes (ARGs) is increasing concern as a serious threat to ecological security and human health. Irrigation with sewage farmland application manure or biosolids in agricultural practices introduce substantial selective agents such antibiotics toxic metals, aggravating the transfer ARGs from soil environment humans via food chain. To address this issue, hyperaccumulator (Sedum plumbizincicola) combined biochar amendment was first used investigate mitigation cadmium oxytetracycline co-contaminated by conducting pot experiment. addition affected distribution plants differently enhancing their but restraining transmission S. plumbizincicola. planting plumbizincicola resulted an increase environment. A structural equation model illustrated that mobile genetic elements played dominant role shaping profile ARGs. Taken together, these findings provide practical understanding for mitigating system complex contamination can have profound significance management regard ARG dissemination control.
