Background: Biomarkers are important tools in toxicology that can improve risk assessment, early detection of organ injury, and monitoring pharmacological responses. However, rigorous validation is necessary to justify use for regulatory decisionmaking. Purpose: This article reviews the current landscape surrounding established emerging biomarkers toxicity across multiple techniques, with a focus on assessment merits, limitations, needs. Main Body: An overview common clinical chemistry used safety liver, kidney, heart, inflammation first provided, highlighting their biological relevance applications detecting dysfunction. Novel protein enhanced sensitivity injury then discussed, such as microRNAs liver clusterin kidney. Metabolomic biomarker approaches assess biofluids mitochondrial outlined, along toxicogenomic markers susceptibility like HLA alleles. Non-invasive imaging techniques including ultrasound, MRI, PET tracers function explored provide additional modalities measurement. Emerging microphysiological systems high-throughput omics discovery also described. Conclusion: While great progress has been made, translation qualification novel remains challenging. review synthesizes key developments categories, evaluates readiness use, outlines strategic considerations fit-for-purpose advance integration into drug development programs.

Single-organic-molecule fluorescent probes with double-lock or even multi-lock response modes have attracted the attention of a wide range researchers. The number corresponding reports has rapidly increased in recent years. effective application mode single-molecule probe improved comprehensive understanding related targets' functions influences pathologic processes. Building highly efficient functional would benefit diagnosis and treatment diseases. Here, we conducted theoretical analysis synthesizing sensing mechanism this kind probe, thereby guiding design building new probes. In work, discuss detail electronic structure, electron excitation, character recently developed which could achieve discrimination profiling spontaneous reactive oxygen species (ROS, •OH, HClO) simultaneously. results provide insights that will help develop tools for biological medical fields.

Abstract Cardiovascular diseases (CVD) represent a major challenge for the elderly population and continue to be among leading causes of mortality worldwide. The integrity mitochondria in peripheral lymphocytes can indicate age-related degenerative disorders, revealing balance between bioenergetics, inflammation, senescence. present study aims investigate impact aging on mitochondrial-mediated epigenetic stress response mechanisms both young individuals. Mitochondrial oxidative DNA damage, repair response, copy number, methylation, fusion, fission genes, electron transport chain enzyme complex activities, pro-inflammatory cytokine levels, associated were studied. results demonstrate that group experienced more significant disruption expression mitochondrial impaired (mtDNA) intricately linked mitochondrial-nuclear crosstalk, increasing their susceptibility CVD. findings highlight mitochondria’s crucial role cellular dynamics, influencing responses through fusion cycles. Overall, these insights provide valuable information about molecular processes linking dysfunction cardiovascular risks, paving way further exploration potential interventions aging-related health outcomes.
