ABSTRACT Gut microbes can impact cognition and behavior, but whether they regulate the division of labor in animal societies is unknown. We addressed this question using honeybees since exhibit between nurses foragers because their gut microbiota be manipulated. Using automated behavioral tracking controlling for co-housing effects, we show that influence age at which bees start expressing foraging-like behaviors laboratory have no effects on time spent a foraging arena number trips. Moreover, did not hallmarks maturation such as body weight, cuticular hydrocarbon profile, hypopharyngeal gland size, gene expression, proportion maturing into foragers. Overall, study shows honeybee plays role onset foraging-related behavior without permanent consequences colony-level several physiological maturation. IMPORTANCE The emerging model system studying microbiota-host interactions. Previous studies reported multiple worker bee phenotypes, all change during maturation—the transition from nursing to foraging. tested documented may stem an effect only subtly affected maturation: it accelerated affecting overall or average output. also found host (CHC) expression maturation-related genes. These results are inconsistent with previous reporting weight CHC profile. Our experiments revealed co-housed tend converge physiology, suggesting spurious associations emerge when rearing environments replicated sufficiently accounted analytically.

Abstract Social insect castes (e.g., queens, workers) are prime examples of phenotypic plasticity (i.e., different phenotypes arising from the same genotype). Yet, mechanisms that give rise to highly fertile, long-lived queens versus non-reproducing, short-lived workers not well understood. Recently, a module co-expressed genes has been identified characterizes compared termite Cryptotermes secundus (Kalotermitidae): Queen Central Module (QCM). We tested whether QCM is shared in species, which gradually develop via early larval and late instars, latter functioning as totipotent (linear development). Similar C. , gene expression profiles revealed an enrichment Zootermopsis angusticollis species another family (Archotermopsidae). The these became enriched during development instars queens. Thus, our results support hypothesis conserved genetic toolkit with gradual linear development. Our data also imply strong caste-specific tissue specificity signal being restricted head-prothorax tissues This tissue-specific key aging-related might have facilitated evolution long lifespan

ABSTRACT Insects display exceptional phenotypic plasticity, which can be mediated by epigenetic modifications, including CpG methylation and histone modifications. In vertebrates, both are interlinked is associated with gene repression. However, little known about these regulatory systems in invertebrates, where mainly restricted to bodies of transcriptionally active genes. A widely conserved mechanism involves the co‐transcriptional deposition H3K36 trimethylation targeted unmethylated CpGs de novo DNA methyltransferase DNMT3. DNMT3 has been lost multiple times invertebrate lineages raising question how links between methylation, modifications expression affected its loss. Here, we report landscape Leptinotarsa decemlineata , a beetle species that but retained methylation. We combine RNA‐seq, enzymatic methyl‐seq CUT&Tag study expression, patterns H3K36me3 H3K27ac on genome‐wide scale. Despite loss DNMT3, mirrors patterns. Together, they give rise signature profiles for expressed not show prominent peak at transcription start site predictive genes irrespective their status. Our provides new insights into evolutionary flexibility modification urge caution when generalizing across species.

The role of epigenetics in regulating caste polyphenism social insects has been debated. Here, we tested the importance histone de/acetylation processes for maintenance queen hallmarks like a high fecundity and long lifespan. To this end, performed RNA interference experiments against deacetylase 3 (HDAC3) termite Cryptotermes secundus. Fat body transcriptomes chemical communication profiles revealed that silencing HDAC3 leads to signals indicative hallmarks. This includes fostering signalling, defence ageing reduction life-shortening IIS (insulin/insulin-like growth factor signalling) endocrine JH (juvenile hormone) signalling via Kr-h1 (Krüppel-homologue 1). These observed patterns were similar those protein-enriched diet, which might imply acetylation conveys nutritional effects. Strikingly, contrast solitary insects, reduced had no negative effect on fecundity-related vitellogenesis fat bodies. suggests an uncoupling longevity pathways from bodies, can help explain queens' extraordinary lifespans combined with fecundity.
