Phosphorus (P) is an important and irreplaceable macronutrient. It central to energy information storage exchange in living cells. ABSTRACT Polyphosphate (polyP) is an intriguing molecule that found in almost any organism, covering a multitude of cellular functions. In industry, polyP used due to its unique physiochemical properties, including pH buffering, water binding, and bacteriostatic activities. Despite the importance polyP, analytics still challenging, with gold standard being 31 P NMR. Here, we present simple staining method using fluorescent dye JC‐D7 for semi‐quantitative evaluation yeast extracts. Notably, fluorescence response was affected by concentration polymer chain length 0.5–500 µg/mL range. Hence, samples unknown compositions, cannot be absolute quantification. Fluorescence unaffected inorganic phosphate up 50 mM. Abstract Inorganic polyphosphate, a linear polymer of orthophosphate residues linked by phosphoanhydride bonds, occurs in all three domains life and plays diverse prominent role metabolism cellular regulation. While the polyphosphate its physiological significance have been well studied bacteria eukaryotes including human, there are only few studies archaea available so far.
