Abstract Gastric cancer (GC) remains a leading cause of cancer-related mortality worldwide, with limited treatment options in advanced stages. Immunotherapy, particularly immune checkpoint inhibitors (ICIs) targeting PD1/PD-L1, has emerged as promising therapeutic approach. However, significant proportion patients exhibit primary or acquired resistance, limiting the overall efficacy immunotherapy. This review provides comprehensive analysis mechanisms underlying immunotherapy resistance GC, including role tumor microenvironment, dynamic PD-L1 expression, compensatory activation other checkpoints, and genomic instability. Furthermore, explores GC-specific factors such molecular subtypes, unique evasion mechanisms, impact Helicobacter pylori infection. We also discuss emerging strategies to overcome combination therapies, novel immunotherapeutic approaches, personalized based on genomics microenvironment. By highlighting these key areas, this aims inform future research directions clinical practice, ultimately improving outcomes for GC undergoing

Abstract Long non-coding RNAs (lncRNAs) have emerged as crucial regulators in various cellular processes, including cancer progression and stress response. Recent studies demonstrated that copper accumulation induces a unique form of cell death known cuproptosis, with lncRNAs playing key role regulating cuproptosis-associated pathways. These may trigger cell-specific responses to stress, presenting new opportunities prognostic markers therapeutic targets. This paper delves into the cuproptosis-mediated cancer, underscoring their potential biomarkers targets for innovative strategies. A thorough review scientific literature was conducted, utilizing databases such PubMed, Google Scholar, ScienceDirect, search terms like 'lncRNAs,' 'cuproptosis,' 'cancer.' Studies were selected based on relevance lncRNA regulation cuproptosis pathways implications prognosis treatment. The highlights significant contribution cuproptosis-related genes pathways, impacting metabolism, mitochondrial responses, apoptotic signaling. Specific are breast, lung, liver, ovarian, pancreatic, gastric cancers. objective this article is explore cancers mediated by cuproptosis.

Gastric cancer represents a highly lethal malignancy with an elevated mortality rate among patients, coupled suboptimal postoperative survival prognosis. Nectin-4, overexpressed oncological target for various cancers, has been exploited to create antibody-drug conjugates (ADCs) treat solid tumors. However, there is limited research on Nectin-4 ADCs specifically gastric cancer, and conventional immunoglobulin G (IgG)-based frequently encounter binding site barriers. Based the excellent tumor penetration capabilities inherent in nanobodies (Nbs), we developed Nectin-4-targeting Nb drug (NDCs) treatment of cancer.

ABSTRACT Chemotherapy is an effective treatment for gastric cancer. However, many patients develop resistance to chemotherapeutic agents during clinical treatment. LncRNA CCAT1 has recently been shown influence cellular specific drugs, but its role in cancer remains underexplored. This study aims investigate the of cisplatin cells and potential underlying mechanisms. Gastric cell lines with acquired were established. The expression was assessed both cisplatin‐sensitive cisplatin‐resistant AGS lines. knocked down AGS/DDP cells, changes IC50 values measured using Cell Counting Kit‐8 (CCK‐8) assay. Apoptosis evaluated by flow cytometry. Additionally, Western blotting employed measure levels PI3K/AKT/mTOR signaling pathway proteins apoptosis‐related interference control groups. RT‐qPCR results indicated that significantly elevated compared non‐resistant cells. Similarly, CCK‐8 assay demonstrated knocking resistant increased their sensitivity Flow cytometry blot further confirmed silencing promoted apoptosis these higher sensitive counterparts, resulted reduced proteins.
