Some issues in pharmaceutical therapies such as instability, poor membrane permeability, and bioavailability of drugs can be solved by the design suitable delivery systems based on combination two pillar classes ingredients: polymers lipids. At same time, modern technologies are required to overcome production limitations (low productivity, high energy consumption, expensive setup, long process times) pass at industrial level. In this paper, a summary applications polymeric lipid materials combined nanostructures (hybrid nanocarriers) is reported. Then, recent techniques adopted hybrid nanoparticles discussed, highlighting still present that hold back implementation.

Chitosan is a product of the deacetylation chitin, which widely found in nature. insoluble water and most organic solvents, seriously limits both its application scope applicable fields. However, chitosan contains active functional groups that are liable to chemical reactions; thus, derivatives can be obtained through modification chitosan. The has been an important aspect research, showing better solubility, pH-sensitive targeting, increased number delivery systems, etc. This review summarizes by acylation, carboxylation, alkylation, quaternization order improve pH sensitivity, targeting derivatives. applications antibacterial, sustained slowly release, system fields also described. will have large impact show potential biomedicine for development drugs future.

Intercellular tight junctions represent a formidable barrier against paracellular drug absorption at epithelia (e.g., nasal, intestinal) and the endothelium blood–brain barrier). In order to enhance transport of drugs increase their bioavailability organ deposition, active excipients modulating have been applied. First-generation permeation enhancers (PEs) acted by unspecific interactions, while recently developed PEs address specific physiological mechanisms. Such target junction modulators (TJMs) advantage defined mechanism action. To date, merely few these novel has entered into clinical trials, as lack in safety efficiency vivo often impedes commercialisation. A stronger focus on development such would result an economic therapeutic improvement current future drugs.
