Determining a novel etiology and mechanism of human cancer requires extraction characteristic mutational signatures derived from chemical substances. This study explored the N-nitroso bile acid conjugates using Salmonella strains. Exposing S. typhimurium TA1535 to N-nitroso-glycine/taurine induced predominance C:G T:A transitions. Two signatures, B1 B2, were extracted. Signature is associated with N-nitroso-glycine conjugates, while B2 linked N-nitroso-taurine conjugates. revealed strong transcribed strand bias GCC GCT contexts, mutation pattern in YG7108, which lacks O6-methylguanine DNA methyltransferases, matched that wild-type strain TA1535, suggesting O6-methyl-deoxyguanosine contributes mutations relevant regions. COSMIC database-based similarity analysis closely resembled SBS42, occupational cholangiocarcinoma caused by overexposure 1,2-dichlolopropane (1,2-DCP) and/or dichloromethane (DCM). Moreover, inflammatory response pathway was 1,2-DCP exposure cholangiocyte cell line, iNOS expression positive cholangiocarcinomas. These results suggest triggers an biliary epithelial cells upregulating conjugate production, resulting via adduct formation.

The transformative role of artificial intelligence (AI) and multiomics could enhance the diagnostic prognostic capabilities liquid biopsy (LB) for lung cancer (LC). Despite advances, transition from tissue biopsies to more sophisticated, non-invasive methods like LB has been impeded by challenges such as heterogeneity biomarkers low concentration tumour-related analytes. advent - enabled deep learning algorithms offers a solution allowing simultaneous analysis various analytes across multiple biological fluids, presenting paradigm shift in diagnostics.
