1 Abstract The diverse functions of different cortical areas are thought to arise from their distinct groups inputs. Visual stimuli that deviate from the current context elicit augmented responses in primary visual cortex (V1). These heightened responses, known as "deviance detection," require local inhibition V1 and top-down input anterior cingulate area (ACa). Here, we investigated mechanisms by which ACa interact to support deviance detection. Local field potential recordings mice during an oddball paradigm showed ACa-V1 synchrony peaks theta/alpha band (≈10 Hz). Two-photon imaging revealed mainly pyramidal neurons exhibited detection, while contextually redundant increased vasoactive intestinal peptide (VIP)-positive interneuron (VIP) activity decreased somatostatin-positive (SST) activity. Optogenetic drive of inputs at 10 Hz activated V1-VIPs but inhibited V1-SSTs, mirroring dynamics present paradigm. Chemogenetic disrupted Aca-V1 detection V1. results outline temporal interneuron-specific modulation processing.

Under the predictive coding framework, brain generates a model of environment based on previous experiences. Incoming sensory information is compared to this model, such that if predictions do not match inputs, prediction error generated. Predictions are passed top-down, and errors emerge when bottom-up does predictions. Prediction occur sequentially in primary auditory cortex (A1) then medial prefrontal (mPFC). Here, we test hypothesis mPFC sends contribute generation errors. We used optogenetics block top-down signals from while recording neuronal A1 under classical "oddball" paradigm. Blocking reduces response rare sounds, it affect responses predictable or random sounds. Our results provide empirical evidence for enhance unpredicted stimuli.

In the field of learning theory and practice, superior efficacy multisensory over uni-sensory is well-accepted. However, underlying neural mechanisms at macro-level human brain remain largely unexplored. This study addresses this gap by providing novel empirical evidence a theoretical framework for understanding superiority learning. Through cognitive, behavioral, electroencephalographic assessment carefully controlled training interventions, our uncovers fundamental distinction in their neuroplastic patterns. A multilayered network analysis pre- post- EEG data allowed us to model connectivity within across different frequency bands cortical level. Pre-training unveils complex distributed sources communicating through cross-frequency coupling, while comparison post-training demonstrates significant differences reorganizational patterns Uni-sensory primarily modifies coupling between lower higher frequencies, whereas induces changes beta band more focused network, implying development unified representation audiovisual stimuli. combination with behavioural cognitive findings suggests that, benefits from an automatic top-down transfer training, relies mainly on limited bottom-up generalization. Abstract Serotonergic psychedelics (e.g., psilocybin) have shown potential for treating psychiatric disorders, with therapeutic effects lasting weeks after a single dose. Predictive processing theories posit that work by loosening priors or high-level beliefs, including ingrained biases become pathological, leading to shifts in bottom-up vs top-down information reconfigure perception, cognition, and mood. Because 5-HT2A receptors, the primary target of psychedelics, are enriched visual cortices, we investigated whether alter manner consistent predictive theories. People who recently (<3 weeks) used 5-HT2A-agonist (psilocybin, LSD) exhibited slowed response latencies increased cortical involvement generating saccades targets predictable locations, along generalization sensory prediction errors (i.e., deviance detection) during passive processing. Individuals 5-HT1A- selective psychedelic (5-MeO-DMT) displayed similar changes saccade production, but unaltered detection, suggesting circuit-specific effects. Mice administered DOI (5- HT2A-agonist) altered detection within cortex (V1), weakened feedback V1 from higher area ACa. These results concord hypothesis shift balance circuits – an effect persists beyond acute exposure period.
