TJs maintain the epithelial barrier by regulating paracellular permeability. Since are under dynamically fluctuating intercellular tension, cells must continuously survey and repair any damage. However, underlying mechanisms allowing to sense TJ damage not yet fully understood. Here, we showed that proteinases play an important role in maintenance of barrier. At break sites, EpCAM-claudin-7 complexes on basolateral membrane become accessible apical membrane-anchored serine (MASPs) MASPs cleave EpCAM. Biochemical data imaging analysis suggest claudin-7 released from EpCAM contributes rapid damaged TJs. Knockout (KO) drastically reduced function live-imaging permeability MASPs-KO exhibited increased size, duration, frequency leaks. Together, our results reveal a novel mechanism through localized proteolysis at leaks, provide better understanding dynamic regulation

Epithelial tissues are dramatically remodelled during embryogenesis and tissue homeostasis yet need to maintain their sealing properties sustain barrier functions at any time. Part of these remodellings involve the elimination a large proportion cells through apoptosis. Cell extrusion, remodelling steps leading seamless dying cell expulsion, helps cohesion. However, there is an intrinsic limit in system that can only accommodate certain proportion/rate as well spatiotemporal distributions. What then critical conditions epithelial rupture/tear/sealing defects upon which mechanisms ensure such limits never reached? In this short review, I document rupture has been observed, including contexts death, mechanical parameters influencing rupture, review feedback help keep epithelia away from breaking point.
