Convergent extension (CE) is a universal morphogenetic engine that promotes polarized tissue extension. The frizzled (fz) and dishevelled (dsh) genes are highly conserved members of both the planar cell polarity (PCP) pathway Wnt signaling pathway. Body axis specification is a crucial event in animal embryogenesis and was an essential evolutionary innovation for founding the kingdom. It involves two distinct components that coordinate to establish spatial organisation of embryo: initiation cascades regionalised gene expression orientation morphogenetic processes such as body elongation. Intense interest first component has revealed Wnt/β-catenin signalling ancestrally responsible initiating regional expression, but origin oriented morphogenesis received little attention. Abstract Planar polarity establishment in epithelia requires interpretation of directional tissue-level information at cellular and molecular levels. Mechanical forces exerted during tissue morphogenesis are emerging as crucial cues, yet the mechanisms by which they regulate planar poorly understood. Using Drosophila pupal wing, we confirm that stress promotes proximal-distal (PD) alignment. Moreover, high anisotropy can reduce rate accumulation lower stability on cell junctions core protein Frizzled (Fz). Notably, under anisotropy, see an increased gradient flow, characterised differential velocities across adjacent rows. This turnover cell-cell contacts parallel to flow direction, possibly via dissociation transmembrane complexes shear forces.
