Radioiodine refractory differentiated thyroid cancer (RAIR-DTC) is difficult to treat with radioactive iodine because of the absence sodium iodide transporter (NIS) in basement membrane follicular cells for uptake. This usually due mutation or rearrangement genes and aberrant activation signal pathways, which result abnormal expression thyroid-specific genes, leading resistance radioiodine therapy. Therefore, inhibiting proliferation growth RAIR-DTC multi-kinase inhibitors (MKIs) other chemicals restoring its differentiation then carrying out therapy have become first-line treatment strategies main research directions. The drugs that regulate these kinases signaling pathways been studied clinical preclinical settings. In this review article, we summarized major gene mutation, rearrangement, led RAIR-DTC, as well agents tested trials.

Thyroid cancer (TC) is the most common endocrine malignancy, with a rapidly increasing global incidence. Scutellariae Barbatae Herba (SBH) exhibits significant antitumor activity; however, its mechanism against TC remains unclear. This study aims to explore immunotherapeutic of SBH in treating through network pharmacology, bioinformatics analysis, and experimental validation. In TCMSP database, active components potential targets were screened construct drug-component-target-disease network. then filtered, selected build protein-protein interaction GO KEGG enrichment analyses performed. The expression, prognosis, roles core genes validated using TCGA databases. Molecular docking demonstrated binding interactions between targets. Finally, vitro experiments conducted validate results pharmacology analysis. 14 29 identified from database. PPI analysis highlighted SPP1 as key target. involved 722 biological processes, 24 cellular components, 73 molecular functions. suggested that anticancer effect might be mediated signaling pathways such AGE-RAGE PI3K-Akt. data indicated highly expressed associated diagnosis, pathological stage, N gender patients. Additionally, expression correlated infiltration types immune cells, highest correlation observed macrophages. has high stability quercetin, Rhamnazin, Salvigenin, energies -8.117, -7.494, − 7.202 kJ∙mol 1, respectively. Experimental validation showed quercetin inhibited growth cells dose-dependent manner. Protein downregulated mRNA protein expression. combines database predictions reveal mechanisms TC, providing effective strategies for immunotherapy TC.

We developed a named entity (NE) framework for information extraction from semi-structured clinical notes retrieved The Cancer Genome Atlas-Thyroid (TCGA-THCA) database and examined Large Language Models (LLMs) strategies to classify the 8th edition of American Joint Committee on (AJCC) staging Thyroid Association (ATA) risk category patients with well-differentiated thyroid cancer. NE consisted annotation guidelines development, ground truth labelling, prompting approaches, evaluation codes. Four LLMs (Mistral-7B-Instruct, Llama-3.1-8B-Instruct, Gemma-2-9B-Instruct, Qwen2.5-7B-Instruct) were offline utilised extraction, comparing expert-curated truth. Our was using 50 TCGA-THCA pathology notes. 289 35 pseudo-clinical cases used validation. Taking an ensemble-like majority-vote strategy achieved satisfactory performance AJCC ATA in both development validation sets. ensemble classifier optimised efficiency accuracy classifying stage cancer patients.

γ-Glutamine cyclotransferase (GGCT) is an enzyme involved in the metabolic cycle of glutathione (GSH). Abnormal GSH metabolism mostly related to ferroptosis. However, mechanisms supporting aberrant GGCT expression PTC remain be investigated. In this study, we found that knockdown inhibited synthesis and promoted malondialdehyde (MDA) reactive oxygen species (ROS) accumulation, thereby promoting ferroptosis papillary thyroid cancer cells. Pro-GA, specific inhibitor GGCT, subcutaneous tumor formation K1 IP combined with LC-MS/MS showed interaction between RPS15A. RPS15A highly expressed tissues cells, promotes stability Knockdown p53 expression, which turn SLC7A11 resulting ferroptosis, while overexpression reversed GGCT-induced addition, miR-205-5p targeted 3' UTR inhibit GGCT-mediated growth, lung metastasis. conclusion, Mechanistically, interacts RPS15A, protein inhibits inhibiting synthesis. The upstream mechanism regulation by targeting GGCT.

Since its inception in 2012, the Nuclear Medicine Global Initiative (NMGI) of Society and Molecular Imaging has played an important role addressing significant challenges field nuclear medicine molecular imaging. The first 3 projects were dedicated to standardizing pediatric practices, global radionuclide access availability, assessing educational training initiatives on theranostics across globe. These efforts aimed advance human health, foster worldwide collaboration, standardize procedural guidelines enhance quality safety practice. In latest project, NMGI develop a unified nomenclature for systemic therapy medicine, diverse terminology currently used. An online survey was distributed member organizations, drawing participation from various geographical locations disciplines. anonymously collected responses physicians, physicists, scientists, radiopharmacists, radiopharmaceutical dosimetrists, technologists, nurse managers, totaling 240 30 countries. Findings revealed prevailing use term targeted therapy, with 52% respondents expressing preference this term. contrast, approximately 37% favored "radiopharmaceutical therapy," whereas 11% "molecular therapy." Other key terms under umbrella also discussed achieve consensus terminology. dynamic fluid should improve communication within field, better reflect technology used, enable comparison results, ultimately lead improved patient outcomes.
