Abstract Allergic rhinitis (AR) is characterized by nasal symptoms such as rubbing and sneezing, often triggered allergen exposure. The purpose of this study to dissect the roles NLRP3‐mediated immune modulation macrophage pyroptosis in modulating T cell differentiation within context ovalbumin (OVA)‐induced AR mice. OVA‐induced was established mice, evaluating symptoms, infiltration, cytokine levels, differentiation. Manipulations using NLRP3−/−, ASC−/− clodronate liposome treatment, NLRP3 inhibitor MCC950 were performed assess their impact on responses. Following OVA stimulation, increased observed group along with augmented GATA3 expression elevated IL‐4 IL‐1b indicative Th2 polarization cellular involvement. NLRP3−/− mice exhibited reduced CD3+ cells post induction, implicating AR. In recent years, there has been a global increase in the prevalence of allergic diseases, including rhinitis, chronic rhinosinusitis, asthma, atopic dermatitis, conjunctivitis, and food allergies. Since pathogenic mechanisms these diseases are not yet fully understood, targeted effective therapies lacking. The NLRP3 inflammasome, multiprotein complex implicated various inflammatory can be activated by diverse stimuli. It assembles into inflammasome complexes through conformational changes, initiating proteolytic cleavage dormant procaspase-1 active caspase-1 promoting maturation cytokines, IL-1β IL-18. Dysfunction may serve as key driver leading to pyroptosis amplifying local response. As preliminarily demonstrated, specific vesicle inhibitors play refectory roles animal models it is believed that potential therapeutic agents for diseases. Plasma cell mastitis (PCM) is a sterile inflammatory condition primarily characterized by periductal inflammation and ductal ectasia. Currently, there lack of non-invasive or minimally invasive treatment option other than surgical intervention. The NLRP3 inflammasome has been implicated in the pathogenesis progression various diseases, however, its involvement PCM not yet reported. In this study, we initially observed pronounced upregulation both human mouse tissue elucidated mechanism underlying attenuation through inhibition NLRP3. We established murine model collected samples on day 14, when reached peak, for subsequent research purposes. MCC950, an inhibitor, was utilized to effectively ameliorate significantly reducing plasma infiltration mammary tissue, as well attenuate expression pro-inflammatory cytokines including IL-1β, TNF-α, IL-2, IL-6. Mechanistically, that MCC950 augmented function myeloid-derived suppressor cells (MDSCs), which turn inhibited cells. Furthermore, it noted depleting MDSCs greatly compromised therapeutic efficacy MCC950. Collectively, our findings suggest administration potential impede augmenting numerically functionally, ultimately treating effectively. This study provides valuable insights into utilization pharmacological agents treatment.
