Interferons (IFNs) stimulate the expression of numerous IFN-stimulating genes via Janus kinase-signal transducers and activators transcription (JAK-STAT) signaling pathway, which plays an important role in host defense against viral infections. In mammals, including humans mice, a substantial number IFN-stimulated (ISGs) have been identified, their molecular mechanisms elucidated. It is to note that avian species are phylogenetically distant from resulting distinct IFN-induced ISGs may different functions. Abstract The human intestinal enteroid (HIE) cell culture system with the support of glycine-conjugated bile acid glycochenodeoxycholic (GCDCA) and ceramide (C2) facilitate successful replication several norovirus strains. Here we investigate how presence GCDCA/C2 impacts gene expression norovirus-infected HIE impact 25 hydroxycholesterol (25-HC), a key regulator cholesterol homeostasis production on replication. Importance Norovirus is leading cause epidemic endemic acute gastroenteritis worldwide currently, there are no effective therapeutic strategies against this highly contagious pathogen. Our study provides insights into highlight role cholesterol/oxysterol replication, demonstrate potential utility oxysterols developing therapeutics.
