Diabetes
mellitus
(DM)

prevalent
chronic
metabolic
diseases,
characterized
primarily
by
a
persistent
elevation
in
blood
glucose
levels.
This
condition
typically
stems
from
either
insufficient
insulin
secretion
or
functional
defect
itself.
Clinically,
diabetes
is
classified
into
type
1
(T1DM)
and
2
(T2DM),
with
T2DM
comprising
nearly
90%
of
all
diagnosed
cases.
Notably,
global
incidence
has
surged
dramatically
over
recent
decades.
The
adenylate-activated
protein
kinase
(AMPK)
signaling
pathway
crucial
regulating
cellular
energy
metabolism,
marking
it
as
significant
therapeutic
target
for
related
complications.
Natural
products,
their
diverse
origins,
multifaceted
bioactivities,
relative
safety,
hold
considerable
promise
modulating
AMPK
pathway.

Acetaminophen
(APAP)-induced
liver
injury
is
a
common
hepatic
disease
resulting
from
drug
abuse.
Few
targeted
treatments
are
available
clinically
nowadays.
The
flower
bud
of
Rosa
rugosa
has
wide
range
biological
activities.
However,
it
unclear
whether
alleviates
caused
by
APAP.
Here,
we
prepared
an
ethanol
extract
(ERS)
and
analyzed
its
chemical
profile.
Furthermore,
revealed
that
ERS
significantly
ameliorated
APAP-induced
apoptosis
ferroptosis
in
AML-12
hepatocytes
dampened
APAP-mediated
cytotoxicity.
In
cells,
elevated
Sirt1
expression,
boosted
the
LKB1/AMPK/Nrf2
axis,
thereby
crippled
intracellular
oxidative
stress.
Both
EX527
NAM,
which
chemically
unrelated
inhibitors
Sirt1,
blocked
ERS-induced
activation
signaling.
protection
against
APAP-triggered
toxicity
cells
was
subsequently
abolished.
As
expression
LKB1
knocked
down,
still
upregulated
but
failed
to
activate
AMPK/Nrf2
cascade
or
suppress
cytotoxicity
provoked
Results
vivo
experiments
showed
attenuated
APAP-caused
hepatocyte
improved
inflammation.
Consistently,
increased
phosphorylations
AMPK,
promoted
Nrf2
nuclear
translocation
livers
APAP-intoxicated
mice.
Hepatic
transcriptions
HO-1
GCLC,
downstream
antioxidant
genes
Nrf2,
were
also
response
ERS.
Our
results
collectively
indicated
effectively
attenuates
activating
cascade.
Upregulated
plays
crucial
role
ERS-mediated
LKB1.
