Background The incidence rate of adolescent depression and anxiety has been increasing since the outbreak COVID-19, which there are no effective therapeutic drugs available. Si-ni San is commonly used in traditional Chinese medicine for treatment depression-like as well anxiety-like behavior, but its mechanism treating combined with during adolescence not yet clear. Methods Network pharmacology was to explore potential drug molecules related targets, molecular docking dynamics (MD) simulation were evaluate interaction between a model rats following behavioral tests biology verify results from network docking. Results As result, 256 active ingredients 1128 targets screened out. Among them, quercetin, Luteolin, kaempferol, 7-Methoxy-2-methyl isoflavone, formononetin showed be most ingredients; while STAT3, IL6, TNF, AKT1, TP53, IL1B, MAPK3, VEGFA, CASP3, MMP9 targets. AGE-RAGE signaling pathway diabetic complications, IL-17 pathway, HIF-1 PI3K-Akt TNF involved anti-inflammation processes, probable pathways regulated by San. Molecular MD compounds inflammation-associated revealed good binding abilities nobiletin PTGS2 PPARγ. In experiment rats, markedly suppressed early maternal separation (MS) chronic unpredictable mild stress (CUMS)-induced anxiety. qPCR further indicated that oxidative inflammatory response. Conclusion This study demonstrates anxiety- behavior induced MS CUMS can ameliorated improved inflammation hippocampus via targeting Nrf2 helping reveal

Phytolacca americana fruits exhibit a wide range of biological activities, including antimicrobial, anti-inflammatory, and anticancer properties. This study aims to investigate the phenolic profile hydroethanolic extracts from both fresh (PEC) dried (PEU) P. using high-performance liquid chromatography (HPLC) evaluate their impact on anxiety-like behavior, memory, oxidative stress, cholinergic status in zebrafish (Danio rerio, Tübingen strain) treated with scopolamine (SCO, 100 μM). Acute administration PEC PEU (0.1, 0.5, 1 mg/L) was conducted for one hour per day. In silico analyses were performed pharmacokinetic characteristics compounds discerned two extracts, platforms such as SwissAdme, Molinspiration, ProToX-III, AdmetLab 3.0, PKCSM, PASS. Anxiety-like behavior memory performance assessed through specific behavioral assays, novel tank test (NTT), light/dark (LD), approach (NAT), Y-maze, object recognition (NOR). Subsequently, activity acetylcholinesterase (AChE) extent stress brain investigated. Our findings suggest that possess anxiolytic effects, alleviating SCO-induced anxiety enhancing cognitive amnesic zebrafish. Furthermore, these demonstrated ability mitigate deficits by inhibiting AChE supporting antioxidant defense mechanisms increased enzymes reduced lipid protein peroxidation. These results highlight potential use fruit managing impairments related dementia conditions.

Sinapic acid (SA) is a phenylpropanoid derivative found in various natural sources that exhibits remarkable versatile properties, including antioxidant, anti-inflammatory, and metal-chelating capabilities, establishing itself as promising candidate for the prevention treatment of conditions affecting central nervous system, such Alzheimer's disease (AD), Parkinson's (PD), ischemic stroke, other neurological disorders. These effects also include neuroprotection epilepsy models, evidenced by reduction seizure-like behavior, cell death specific hippocampal regions, lowered neuroinflammatory markers. In AD, SA enhances memory, reverses cognitive deficits, attenuates astrocyte activation. has positive on cognition improving memory lowering oxidative stress. This shown lower levels stress markers, higher antioxidant enzyme activity, better retention. Additionally, stroke PD provides microglial protection exerts anti-inflammatory effects. review emphasizes SA's multifaceted neuroprotective properties its potential role brain Despite need further research to fully understand mechanisms action clinical applicability, stands out valuable bioactive compound ongoing quest combat neurodegenerative diseases enhance quality life affected individuals.

Depression is commonly associated with gastrointestinal (GI) disorders, such as constipation, which can potentially intensify depressive symptoms. The interplay between these conditions believed to be facilitated by the gut-brain axis, suggests a complex bidirectional interaction. Current treatments, antidepressants and prokinetics, are often side effects high recurrence rates, highlighting need for effective treatments targeting both depression constipation. This study was designed assess therapeutic efficacy of electroacupuncture (EA) in conjunction Tongbian decoction (TB) management while also investigating underlying mechanisms through lens axis. Chronic unpredictable mild stress (CUMS) employed induce comorbidity model constipation mice, followed administration EA, EA + TB, fluoxetine (FLX). findings demonstrated that antidepressant combination were more pronounced than those alone. TB treatment significantly ameliorated anxiety-like behaviors, restored cognitive function, enhanced motility chronic mice. Furthermore, reduced intestinal inflammation, neuronal morphology, increased expression tryptophan hydroxylase 2 (TPH2) prefrontal cortex (PFC) colon, elevated serum levels 5-hydroxytryptophan (5-HTP)-a molecule acts connector-and promoted synthesis production serotonin (5-HT) tract brain. Contrastingly, FLX showed limited improving In conclusion, regulates TPH2/5-HT pathway via demonstrating synergistic regulation nervous systems, favorable prokinetic effects.
