Background/Objectives: Obesity is a major health concern that can lead to various chronic diseases. Little known about the anti-obesity effect of standardized hot water extract from stems Ipomoea batatas (WIB). This study aimed evaluate therapeutic potential WIB as natural alternative conventional treatments by assessing its effects on body weight, fat accumulation, and key metabolic biomarkers in high-fat diet-induced obesity model. Methods: A diet (HFD) induced C57BL/6 mice. The mice were then treated orally with either orlistat (positive control) or WIB. Changes food intake, weight measured, along blood lipid profiles adipokines. Western blot analyses conducted determine protein levels each tissue. H&E staining white adipose tissue liver, gut microbiota composition analyzed. In brown tissue, enhanced AMPK phosphorylation upregulated thermogenic-related proteins, including peroxisome proliferator-activated receptor-gamma coactivator-1α (PGC-1α), receptor (PPARα), sirtuin 1 (SIRT1), uncoupling (UCP-1), cytochrome C oxidase subunit 4 (COX-IV). Analysis normalized β-diversity reversed HFD-induced phyla imbalances (notably Bacteroidetes, Firmicutes, Proteobacteria). Conclusions: By reducing adiposity under conditions tested murine model, improving markers, favorably modulating microbiota, demonstrates mitigating obesity-related risks.
