The neurotransmitter acetylcholine (ACh) plays a pro-carcinogenic role in various cancer types, including prostate (PCa). existing body of knowledge concerning the mechanisms that underpin protumoral ACh PCa is limited. Glyoxalase 1 (Glo1) metabolic enzyme removes methylglyoxal (MG), an endogenous post-translational modification agent, generating 5-hydro-5-methylimidazolone (MG-H1). Glo1/MG-H1 axis involved tumorigenesis and progression. By using LNCaP PC3 cells, representing extensively studied cell models poorly aggressive bone metastasis-derived PCa, respectively, we found specifically sustains migration, invasion proliferation through Glo1-dependent MG-H1 accumulation with involvement osteopontin (OPN), thus providing novel mechanism underlying ACh’s cells. findings this study unveil hitherto unidentified implicated progression which initiated by involves both OPN. This discovery provides basis for new avenues vivo investigation into physiological relevance roles ACh-driven OPN further research aimed at exploring ways managing progression, aim preventing disease from becoming incurable.
