Abstract Figure G protein-coupled receptors (GPCRs) are central mediators of cell signaling and physiological function. Despite their biological significance, GPCRs have not been widely studied in the field toxicology. Herein, we investigated these novel targets plastic chemicals using a high-throughput drug screening assay with 126 human non-olfactory GPCRs. In first-pass screen, tested activity triphenol phosphate, bisphenol A, diethyl phthalate as well three real-world mixtures extracted from food packaging covering all major polymer types. We found 11 GPCR-chemical interactions, which chemical exhibited most robust at Adenosine receptor 1 (ADORA1) Melatonin (MTNR1A) confirmatory secondary screen. further confirmed that polyvinyl chloride polyurethane products contain ADORA1 or MTNRA1 agonists pharmacological knockdown experiments. Finally, an analysis associated gene ontology terms suggests MTNR1A activation may be linked to downstream effects on circadian metabolic processes. In response to regulatory demands for sustainable practices, there has been a significant increase in the use of recycled materials food packaging, particularly when incorporated behind functional barriers. This study, conducted as part "ABA Modeling" project, evaluates migration contaminants from non-decontaminated, polyethylene terephthalate (rPET) B-layer, which is sandwiched between two virgin A-layers three-layer trays. The study aims assess long-term implications safety within context European and French circular economy frameworks. Using modeling approach based on "worst-case" scenarios ten – similar methodologies employed decontaminated direct contact PET levels chemical residues are analyzed compared. Ageing is a nature process of microplastics that occurrs daily, and human beings are inevitably exposed to aged microplastics. However, systematic understanding ageing status its toxic effect currently still lacking. In this study, plastic cup lids-originated polypropylene (PP) were UV-photoaged until the carbonyl index (CI), canonical indicator for ageing, achieved 0.08, 0.17, 0.22 0.28. The adverse hepatic these PPs (aPPs) was evaluated in Balb/c mice (75 ng/mL water, about 200 particles/day) human-originated liver organoids (LOs, 50 particles/mL, ranged from 5.94 13.15 ng/mL) at low-dose equivalent exposure level. Low-dose PP could induce reductive stress both vitro vivo, by elevating NADH/NAD+ratio CI-dependent manner, together with hepatoxicity (indicated increased AST secretion cytotoxicity), disrupted genes encoding nutrients transporters NADH subunits accompanied restricted ATP supply, declined mitochondrial membrane potential complexI/IV activities, without significant increase MDA levels liver. These changes metabolism, representing circulatory panel increases lactate, triglyceride, Fgf21 levels, decreases pyruvate level, linked body weight gain but elevated contents following aPPs exposure. Additionally, assessing LOs, it found digestion drastically accelerated worsen energy supply upon mitochondria, "scattergun effect" induced formation micro- nano-plastics mixture toward NADH/NAD+imbalance.
