Background & Objective Selenium was one of the essential trace elements that played a pivotal role in human health. Although previous studies have investigated relationship between selenium and non-alcoholic fatty liver disease (NAFLD) fibrosis, these findings were still inconclusive. Our study aimed to explore association blood level NAFLD advanced fibrosis diagnosed by vibration controlled transient elastography (VCTE) US adults. Methods All data extracted from National Health Nutrition Examination Survey database (2017–2018). Participants divided into four groups according quartile level. Liver stiffness attenuation parameter (CAP) measured VCTE. Multiple logistic regression models subgroup analyses conducted determine variety methods. Results A total 3336 participants enrolled main analysis. In multiple models, higher (>205.32, ≤453.62 μg/L) had significant positive with (β = 1.31). Moreover, high significantly inversely 0.61). analysis, correlation found males Despite dietary intake being adjusted or different subgroups, associations NAFLD/advanced remained significant. Conclusions This showed positively among population. lower percentage fibrosis. Blood is more likely cause due imbalances homeostasis rather than intake.Key messagesHigh elastography.Participants fibrosis.NAFLD are caused an imbalance homeostasis, not intake.

This review addresses the role of essential trace element, selenium, in type-2 diabetes mellitus (T2DM) and its metabolic co-morbidities, i.e., syndrome, obesity non-alcoholic fatty liver disease. We refer to dietary requirements selenium key physiological roles selenoproteins. explore dysregulated fuel metabolism T2DM emphasizing relevance inflammation oxidative stress. describe epidemiology observational experimental studies related conditions, explaining that interaction between status glucose control is not limited hyperglycemia but extends hypoglycemia. propose association high plasma/serum T2DM/fasting plasma observed many cross-sectional may rely on upregulation hepatic selenoprotein-P biosynthesis conditions insulin resistance. While animal have revealed potential molecular mechanisms underlying adverse effects severe selenium/selenoprotein excess deficiency pathogenesis resistance β-cell dysfunction, their translational significance rather limited. Importantly, supplementation does appear be a major causal factor for development humans though we cannot currently exclude small contribution top other risk factors, particular if it ingested at (supranutritional) doses. Elevated biomarkers are often measured patients more likely consequence, than cause, diabetes.

Background Systemic immune-inflammation index (SII) is a novel inflammatory marker, and inflammation has been reported to be related with renal damage. We aimed investigate the possible relationship between SII albuminuria. Methods The present cross-sectional study was conducted among adults complete data about urinary albumin-to-creatinine ratio (ACR) in 2005–2018 National Health Nutrition Examination Survey (NHANES). calculated as platelet count × neutrophil count/lymphocyte count. Albuminuria defined ACR >30mg/g. Weighted multivariable regression analysis subgroup were explore independent Results A total of 36,463 individuals included our analysis; 9.56% participants categorized having albuminuria overall increased higher tertiles (tertile 1, 7.83%; tertile 2, 8.49%; 3, 12.13%; p for trend <0.0001). Multivariable logistic showed that level associated likelihood independently (OR = 1.31; 95% CI, 1.17–1.48, p<0.0001) after full adjustment. Subgroup interaction test there no significant dependence gender, age, body mass index, hypertension, diabetes, non‐alcoholic fatty liver disease, estimated glomerular filtration rate (eGFR) on this positive association (all >0.05). Conclusions positively albumin excretion US adults. Further large-scale prospective studies are still needed analyze role

Non-alcoholic fatty liver disease (NAFLD) is a chronic worldwide. Numerous evidence has demonstrated that metabolic reprogramming serves as hallmark associated with an elevated risk of NAFLD progression. Selenoprotein W (SelW) extensively expressed hepatic selenoprotein plays crucial role in antioxidant function. Here, we first SelW significantly distinct factor the tissue patients through Gene Expression Omnibus (GEO) database. Additionally, loss alleviated steatosis induced by high-fat diet (HFD), and was accompanied regulation inflammatory pathways verified transcriptomic analysis. Moreover, co-immunoprecipitation (CO-IP), liquid chromatography-tandem mass spectrometry (LC-MS), laser scanning confocal microscopy (LSCM) molecular docking analysis were subsequently implemented to identify Pyruvate Kinase M2 (PKM2) potential interacting protein SelW. Meanwhile, modulated PKM2 translocation into nucleus trigger transactivation HIF-1α, further mediating mitochondrial apoptosis, eventually resulting damage, ROS excessive production mtDNA leakage. mito-ROS accumulation activation NLRP3 inflammasome-mediated pyroptosis, thereby facilitating extracellular leakage mtDNA. The escaped then evokes cGAS-STING signaling pathway macrophage, thus inducing shift macrophage phenotype. Together, our results suggest promotes hepatocyte apoptosis pyroptosis regulating activate cGAS/STING macrophages, exacerbating progression NAFLD.

Selenium (Se), in the form of selenoproteins, is an essential micronutrient that plays important role human health and disease. To date, there are at least 25 selenoproteins humans involved a wide variety biological functions, including mammalian development, metabolic progress, inflammation response, chemoprotective properties, most notably, oxidoreductase functions. In recent years, numerous studies have reported low Se levels associated with increased risk, poor outcome, mortality disorders, mainly related to limited antioxidant defense resulting from deficiency. Moreover, correlation between deficiency Keshan disease has received considerable attention. Therefore, supplementation as therapeutic strategy for preventing occurrence, delaying progression, alleviating outcomes some diseases been widely studied. However, supranutritional serum may adverse effects, poisoning. This review evaluates status health, particular emphasis on benefits shedding light clinical treatment.
