While cisplatin is an effective anti-tumor treatment, it induces ototoxicity through mechanisms involving DNA damage, oxidative stress, and programmed cell death. Rho-associated coiled-coil-containing protein kinase (ROCK) essential for numerous cellular processes, including apoptosis regulation. Studies have suggested that ROCK inhibitors could prevent promote regeneration. We aimed to investigate the protective effects of inhibitor fasudil against cisplatin-induced in a zebrafish model. The larvae were exposed 1 mM alone or co-administered with varying concentrations 4 h. surviving hair counts, apoptosis, reactive oxygen species (ROS) levels, mitochondrial membrane potential (ΔΨm), caspase 3 activity, autophagy activation assessed. Rheotaxis behavior was also examined. Cisplatin reduced counts; increased ROS production, ΔΨm loss; activated autophagy. Fasudil (100 500 µM) mitigated loss, inhibited activation. preserved by co-administration cisplatin. zebrafish, whereas promising agent ototoxicity.
