Sulfur mustard gas (SM) is a vesicating and alkylating agent used as chemical weapon in many mass-casualty incidents since World War I. Ocular injuries were reported >90% of exposed victims. The mechanisms underlying SM-induced blindness remain elusive. This study tested the hypothesis that corneal fibrosis occurs due to generation myofibroblasts from resident fibroblasts via SMAD2/3 signaling pathway rabbit eyes vivo primary human (hCSFs) isolated donor corneas vitro. Fifty-four New Zealand White Rabbits divided into three groups (Naïve, Vehicle, SM-Vapor treated). group was SM at 200 mg-min/m3 for 8 min MRI Global facility. Rabbit collected on day 3, 7, 14 immunohistochemistry, RNA, protein lysates. caused significant increase SMAD2/3, pSMAD, ɑSMA expression corneas. For mechanistic studies, hCSFs treated with nitrogen (NM) or NM + SIS3 (SMAD3-specific inhibitor) 30 m, h, 24 48 72 h. significantly increased TGFβ, pSMAD3, levels. On contrary, inhibition by treatment reduced hCSFs. We conclude appears play vital role myofibroblast formation cornea following exposure.

Corneal fibrosis is a leading cause of blindness in mammalian species and may result compromised performance sports daily functions. This study evaluated the safety anti-fibrotic effects FDA-approved drugs, angiotensin-converting enzyme inhibitor (ACE-I) lisinopril rho-kinase (ROCK-I) fasudil, alone combination, on canine cornea using an established vitro model.
