Abstract Di-fructofuranose 1,2′:2,3′ dianhydride (DFA-III) is a cyclic fructo-disaccharide, which produced by the condensation of two fructose molecules via caramelization or enzymatic reaction inulin fructotransferase. A strain Blautia producta was known to utilize DFA-III as carbohydrate source; however, mechanisms remain unclear. In this study, we characterized glycoside hydrolase (GH) family 91 (DFA-IIIase) from B. parvula NBRC 113351. Recombinant Bp DFA-IIIase catalyzed reversible conversion inulobiose, further degraded cooperative action and GH32 β- d -fructofuranosidase. utilized in several species with gene cluster for degradation (e.g., 113351, hydrogenotrophica JCM 14656, wexlerae 35486), but not 31267, does possess cluster. Furthermore, hansenii 14655, cannot metabolize fructose, could DFA-III; it degrade presence DFA-III-degrading enzymes. Fecal fermentation tests showed that are important gut microbe degrading DFA-III.
