Legume seed protein is an important source of nutrition, but generally it less digestible than animal protein. Poor digestibility in legume seeds and seedlings may partly reflect defenses against herbivores. Protein changes during germination typically increase proteolysis digestibility, by lowering the levels anti-nutrient protease inhibitors, activating proteases, breaking down storage proteins (including allergens). Germinating sprouts also show striking increases free amino acids (especially asparagine), their roles host defense or other processes are not known. Plant-based proteins are increasingly recognized for their environmental, ethical, and nutritional benefits. However, digestibility varies due to factors such as molecular structure, amino acid composition, processing methods. This review comprehensively analyzes methods used assess plant protein digestibility, including in vivo, vitro, ex vivo approaches. While studies, particularly those using pigs, considered the gold standard, vitro models offer cost-effective reproducible alternatives simulating digestion. Additionally, antinutritional present can hinder necessitating strategies fermentation, enzymatic hydrolysis, high-pressure treatments enhance bioavailability. Advances evaluation techniques, Digestible Indispensable Amino Acid Score (DIAAS) dynamic digestion models, more precise assessments of quality. By systematically comparing these methods, this aims guide food scientists manufacturers selecting appropriate improve quality plant-based products. Abstract Purpose of Review The globally rising food allergy prevalence is associated with the urgent need for new disease prevention methods, efficient treatment, and reliable risk assessment methods characterization allergens. The use of enzymes from the brush border membrane (BBM) in simulated gastrointestinal digestion milk proteins has been evaluated. Brush border membrane (BBM) enzymes greatly affect the bioaccessibility and bioavailability of food nutrients. Despite their physiological importance, a step simulating final stage intestinal digestion has not yet been included in harmonized protocols for vitro digestion, primarily due to challenges replicating dynamics degradation. Herein, we propose an advancement toward more physiologically relevant method, complementing static gastric-duodenal INFOGEST model with missing small phase. BBM hydrolase activity, incubation time, at pH 7.2 were established reproduce conditions. Skim milk powder, as protein food, was subjected digestion. Immediately after duodenal phase, digesta supplemented vesicles purified from pig jejunum.
