Cataracts are primarily caused by aging or gene mutations and the leading cause of blindness globally. As older population increases, number patients with a cataract is expected to grow rapidly. At present, surgery replace lens an artificial intraocular principal treatment method. However, has several drawbacks, including economic burdens complications such as inflammation, xerophthalmia, macular edema posterior capsular opacification. Thus, developing effective non‑surgical strategy beneficial both public health. Abstract Physiochemical tissue inducers and mechanical stimulation are both efficient variables in cartilage fabrication regeneration. In the presence of biomolecules, decellularized extracellular matrix (ECM) may trigger enhance stem cell proliferation differentiation. Here, we investigated controlled release transforming growth factor beta (TGF-β1) as an active mediator mesenchymal stromal cells (MSCs) a biocompatible scaffold for engineering. ECM-derived hydrogel with TGF-β1-loaded alginate-based microspheres (MSs) was created to promote human MSC chondrogenic development. Ex vivo explants complicated multiaxial loading bioreactor replicated physiological conditions. Hydrogels with/without MSs TGF-β1 were highly cytocompatible. MSCs containing TGF-β1/MSs showed comparable gene expression levels those hydrogels added culture media or without TGF-β1. Seventeen novel polyhydroquinoline (1) based Schiff bases (4-20) were synthesized in excellent yields through Hantzsch reaction, characterized by means of spectroscopic techniques and finally screened for their vitro tyrosinase inhibitory potential. Among the series, five compounds 12 (IC50 = 8.93 ± 0.27 µM), 8 16.64 0.32 5 17.74 0.34 4 18.46 0.37 µM) 18 18.64 0.39 found potent inhibitors enzyme comparing with kojic acid 18.30 0.41 µM). Likewise, eight derivatives 7, 15, 9, 6, 10, 13, 14 11 ascribed significant activity having IC50 values 20.72 0.45, 22.53 0.61, 24.17 0.40, 25.72 0.60, 26.43 0.39, 27.90 0.53, 28.37 0.59 29.15 0.64 µM respectively, while remaining four hydrazone-Schiff 19, 20, 17 16 exhibited good to less potential range from 31.58 0.67 63.69 1.30 µM. Docking studies indicate binding hydrazide moiety most active compound site residues tyrosinase. The molecular dynamic simulation indicates that showed root mean square deviation profile complexes stabilized at low nanoseconds, indicating stable conformations.
