Dimethylmonothioarsinic acid (DMMTAV), a potent toxic metabolite of arsenic, exhibits higher cytotoxicity than other arsenicals. This study investigates its influence on NAD(P)H:quinone oxidoreductase (NQO1) regulation in C57BL/6 mice and Hepa-1c1c7 cells. Mice were administered DMMTAV (6mg/kg, IP) with or without TCDD (15µg/kg, IP), hepatic extrahepatic tissues analyzed for NQO1 expression. In vitro, cells treated 0-2µM the presence absence (1nM), levels assessed over time. Western blot, real-time PCR, ARE-luciferase assays determined protein transcriptional regulation. upregulated liver induced time-dependent increase peaking at 12hours. It enhanced TCDD-induced expression increased nuclear NRF2 AHR levels, peak accumulation two hours. activity confirmed activation. These findings reveal enhances primarily via NRF2/AHR pathway activation, providing insight into cellular responses to thioarsenicals.

Cadmium (Cd) is a widely distributed environmental heavy metal pollutant. It extremely toxic to the kidney. This study investigated potential mechanisms of action lentinan (LNT), fungal polysaccharide, on protecting against Cd-induced kidney injury in mice. Male Kunming mice were administered with CdCl2 (2.5 mg/kg/b.w.) by intragastric gavage and LNT drinking water (1 mg/mL) for 10 weeks. Histological examination revealed that reduced glomerular atrophy, lymphocyte infiltration, tubular congestion, collagen accumulation caused Cd exposure. However, oral administration decreased levels promoting excretion feces increasing production metallothionein (MT) In addition, treatment alleviated excessive mitophagy upregulating silencing information regulator1 (SIRT1) prevented subsequent oxidative stress inflammatory responses nuclear factor erythroid 2-related 2 (Nrf2) downregulating kappa-B (NF-κB) signaling pathways. Further, protein expression profibrotic factors, including Tgf-β1, alpha smooth muscle actin, type I 1 chain, progression fibrosis, significantly kidneys treated LNT. Collectively, our findings suggest can relieve nephrotoxicity decreasing its via MT synthesis regulating SIRT1/Nrf2/NF-κB pathway.
