Abstract Studies on insecticide-induced stimulatory effects in pest insects have become of utmost importance due to their potential implications management. Temperature influences the physiology and biology ectothermic organisms like insects. It can also affect toxicity relative efficacy insecticides that are used agricultural fields. The anthranilic diamide insecticide chlorantraniliprole has been extensively applied to control Lepidoptera pests. However, its overuse leads the development of resistance and accumulation residue in environment. Four P450s (CYP6CV5, CYP9A68, CYP321F3, CYP324A12) were first found be constitutively overexpressed an SSB CAP-resistant strain. It is imperative further elucidate molecular mechanisms underlying P450s-mediated CAP for mitigating environmental contamination. Here, we heterologously expressed these four insect cells evaluated their abilities metabolize CAP. Western blotting reduced CO difference spectrum tests showed that P450 proteins had successfully Sf9 cells, which are indicative active functional enzymes. recombinant CYP6CV5, CYP324A12 exhibited a preference metabolizing fluorescent model probe substrates EC, BFC, EFC, EC with enzyme activities 0.54, 0.67, 0.57, 0.46 pmol/min/pmol P450, respectively. In vitro metabolism revealed distinct metabolic rates (0.97, 0.86, 0.75, 0.55 P450) efficiencies (0.45, 0.37, 0.30, 0.17) enzymes, thereby elucidating different protein catalytic activities. Abstract Background The corn leafhopper, Dalbulus maidis , is an important pest in Brazil. While chemical control has traditionally been the cornerstone for managing this pest, field failures have reported some insecticides. To understand if these are due to resistance, we evaluated susceptibility of 11 field‐collected populations D. major insecticides during 2021–2022 crop seasons Brazil using concentration‐mortality bioassays. Additionally, employed diagnostic concentration bioassays and foliar sprays at label‐recommended rates 8–10 collected 2022–2023 seasons. Results High methomyl, carbosulfan acephate was observed on across all tested with resistance ratio (RR) based LC 50 <10‐fold, except one population from Bahia State that exhibited reduced methomyl (RR = 17.5). On other hand, bifenthrin, acetamiprid, imidacloprid, RR ranging 90 2000‐fold. This neonicotinoid pyrethroid further confirmed concentrations 99 susceptible strain, survival >20% mortality <80%. Most exposed low (<5%) high (>80%). Conclusions likely explain controlling study represents first large‐scale monitoring insecticides, results will contribute decision‐making regarding management pest. © 2024 Society Chemical Industry.
