Proanthocyanidins (PACs) have been proven to exert antioxidant and anti-inflammatory effects. In this study, ultra-performance liquid chromatography (UPLC) coupled with linear ion trap-Orbitrap (LTQ-Orbitrap) high-resolution mass spectrometry was first employed systematically screen PACs from the roots of Ephedra sinica Stapf, its ethyl acetate extract (ERE) found contain PAC monomers A-type dimeric proanthocyanidins, which were tentatively identified through characteristic fragmentation patterns. vitro, activity ERE tested 2,2-diphenyl-1-picrylhydrazyl (DPPH) 2, 2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assays. addition, could inhibit production nitric oxide (NO) in lipopolysaccharide (LPS)-induced RAW 264.7 cells. vivo, preventative effects on dextran-sulfate-sodium-induced ulcerative colitis mice investigated. The results showed that alleviated DSS-induced colitis, characterized by decreases disease index (DAI) scores, spleen colon levels TNF-α IL-6, mitigation pathological damage oxidative stress increases length IL-10 levels. conclusion, supplementation derived may offer a new strategy Moreover, our research will greatly facilitate better utilization plants.

Abstract Ulcerative colitis (UC) is currently the most common inflammatory bowel disease (IBD). Due to its diverse and complex causes, there no cure at present, researchers are constantly exploring new therapies. In recent years, nano-selenium particle(SeNP) has attracted wide attention due excellent biological activities. Therefore, in this study, for first time, we used a natural polysaccharide, Eucommia ulmoides polysaccharide (EUP), modified SeNP get EUP-SeNP with size of about 170 nm, effect on 3% dextran sulphate sodium (DSS) induced was explored. Our results showed that colon intestinal histology, mucosal barrier, cytokines microbiome composition were changed after treatment mice. Specifically, it also shown oral could relieve degree DSS-induced mice by restoring weight loss, reducing activity index (DAI), enhancing antioxidant capacity regulating composition. addition, verified mechanism epithelial cell lines, showing inhibited LPS-induced activation TRL-4/NF-κB signaling pathway lines. To some extend, our study provides therapeutic reference IBD.

Background: An animal model of DSS-induced UC has been widely used in basic research, and the dysbiosis gut microbiome is one important pathogenetic mechanisms UC, but its dynamic changes correlation with inflammatory factors are not clear yet. Scope Changes in composition of intestinal microbes may disrupt the balance their interaction with a susceptible host, resulting development inflammatory bowel disease (IBD). Methods and results The study applied combination two Lactobacillus strains ( L. rhamnosus BY‐02, plantarum BY‐05) (“LS treatment”), previously isolates from feces healthy human infants, mouse model dextran sodium sulfate (DSS)‐induced colitis, evaluates ameliorative effect its possible mechanism. LS treatment suppresses weight loss colon shortening, reduces activity index mice. It also has several additional beneficial effects: i) maintains goblet cell numbers ameliorates barrier damage colonic tissue; ii) alters microbial close to normal by increasing abundances Muribaculaceae , Akkermansia Clostridia Oscillospiraceae Lachnospiraceae decreasing abundance Escherichia‐Shigella ; iii) increases content short‐chain fatty acids; iv) pro‐inflammatory lipopolysaccharides; v) overactivation TLR4/NF‐κB signaling pathway. The fungal microbiota may be involved in the regulation of cognition and behavior, while role probiotic fungi against cognitive impairment is unclear. Effects of Clostridium butyricum and chitooligosaccharides (COS), singly in synbiotic combination, were evaluated a C57BL/6 mouse model dextran sulfate (DSS)-induced acute ulcerative colitis (UC). Treatment with C. and/or COS ameliorated UC symptoms vivo, the strongest effects observed for combination terms reduced mortality rates disease activity indices, increased body weight colon length, improved histological features. The achieved following: (i) regulated levels inflammation-related cytokines (tumor necrosis factor alpha [TNF-α], interleukin-1β [IL-1β], IL-6, IL-10) had stronger anti-inflammatory effect than either component alone, based on inhibition Toll-like receptor 4 (TLR-4)/NF-κB/MAPK signaling pathway activation; (ii) enhanced intestinal barrier function by restoring tight junction proteins (occludin, claudin-1, ZO-1) MUC2; (iii) abundance diversity beneficial bacteria (gut microbiota) pathogenic bacteria; (iv) production short-chain fatty acids. Our findings indicate that has strong potential as therapeutic adjuvant UC.
