Abstract Ulcerative colitis (UC) is associated with the shedding of gut mucus. Decellularized porcine small intestinal submucosa (SIS‐ECM) contains numerous active forms extracellular matrix as well growth factors. Herein, a mucus‐mimetic hydrogel (MAG) designed barrier therapy by integrating SIS‐ECM hyaluronic acid and epigallocatechin‐3‐gallate. MAG not only presented viscoelastic behaviors G' 4000 Pa but also good thixotropy. Moreover, physical shielding effective to hinder E. coli invasion. Besides, repairing effect on injured Caco‐2 cells MAG. When biomimetic mucus rectally administrated mouse, it alleviated effectively symptoms improved colonic shorting. expression IL‐1β, IL‐6, TNF‐α at laminae propria substantially inhibited epitheliums are rearranged, tight junction proteins (ZO‐1, Occludin‐1, Claudin‐5) between them greatly upregulated after treatment. Meanwhile, secretion goblets repaired The mechanism mucosa barriers promoting proliferation differentiation stem cells. Collectively, might be promising strategy for UC.

Intestinal microbiota dysbiosis is a well established characteristic of ulcerative colitis (UC). Regulating the gut an effective UC treatment strategy. Berberine (BBR), alkaloid extracted from several Chinese herbs, common traditional medicine. To establish efficacy and mechanism action BBR, we constructed model using healthy adult shorthair cats to conduct systematic study colonic tissue pathology, inflammatory factor expression, structure. We investigated therapeutic capacity BBR for regulating thus work against in 16S rRNA genes amplicon sequencing technology. Our results revealed that dextran sulfate sodium (DSS)-induced cat models showed weight loss, diarrhea accompanied by mucous blood, histological abnormalities, shortening colon, all which were significantly alleviated supplementation with BBR. A gene-based analysis demonstrated could benefit microbiota. Western blot, quantitative PCR, enzyme-linked immunosorbent assays (ELISAs) DSS-induced models, expression factors was increased, activating JAK2/STAT3 signaling pathway, reversed this effect. The myosin light chain (MLC) phosphorylation smooth muscle intestines associated motility inflammation-related cats.
