Recent studies have demonstrated that Sirtuin-1 (SIRT-1)-activating molecules exert a protective role in degenerative ocular diseases. However, these hardly reach the back of eye due to poor solubility aqueous environments and low bioavailability after topical application on eye’s surface. Such hindrances, combined with stability issues, call for need innovative delivery strategies. Within this context, development self-nanoemulsifying drug systems (SNEDDS) SIRT-1 can represent promising approach. The aim work was design optimize SNEDDS two natural agonists, resveratrol (RSV) melatonin (MEL), potential implications treating diabetic retinopathy. Pre-formulation were performed by Design Experiment (DoE) approach construct ternary phase diagram. optimization carried out using Response Surface Methodology (RSM). Ocular drug delivery poses unique challenges due to the complex anatomical and physiological barriers of eye. Conventional dosage forms often fail achieve optimal therapeutic outcomes poor bioavailability, short retention time, off-target effects. In recent years, vesicular systems have emerged as promising solutions address these challenges. Vesicular systems, such liposome, niosome, ethosome, transfersome, others (bilosome, transethosome, cubosome, proniosome, chitosome, terpesome, phytosome, discome, spanlastics), offer several advantages for ocular delivery. These include improved prolonged time on surface, reduced systemic side effects, protection drugs from enzymatic degradation dilution by tears. A total of 16 novel carboxymethyl chitosan derivatives bearing quinoline groups in four classes were prepared by different synthetic methods. Their chemical structures confirmed Fourier-transform infrared spectroscopy (FTIR), nuclear magnetic resonance (NMR), and elemental analysis. The antioxidant experiment results vitro (including DPPH radical scavenging ability, superoxide anion hydroxyl ferric reducing power) demonstrated that adding to (CS) (CMCS) enhanced the ability CS CMCS. Among them, both N, O-CMCS N-TM-O-CMCS showed over 70%. In addition, their radicals reached more than 90% at maximum tested concentration 1.6 mg/mL. Moreover, cytotoxicity assay was carried out on L929 cells MTT method, indicated all no (cell viability > 75%) except derivative 1a, which low 1000 μg/mL 50.77 ± 4.67%). conclusion, remarkable weak cytotoxicity, highlighting potential use food medical applications.
