Abstract BACKGROUND The present study aimed to comprehensively evaluate the anticancer, anti‐inflammatory, and antioxidant properties of Globularia cordifolia L. samples. plant material was collected extracted using maceration method. Antioxidant activities were assessed through DPPH (i.e. 2,2‐diphenyl‐1‐picrylhydrazyl) iron chelation assays, whereas phenolic flavonoid contents measured spectrophotometric methods. Gas chromatography‐mass spectrometry analysis performed identify biologically active compounds. cytotoxic effect on colon cancer cells evaluated MTT [i.e. 2,3‐bis‐(2‐methoxy‐4‐nitro‐5‐sulfophenyl)‐2H‐tetrazolium‐5‐carboxanilide] inflammatory cytokine levels an enzyme‐linked immunosorbent assay. Molecular docking studies investigated interactions bioactive compounds with 4UYA protein, ADME absorption, distribution, metabolism, excretion toxicity) modeling predicted their toxicity. RESULTS revealed that extract exhibited strong activity a median 50% inhibitory concentration (IC 50 ) value 39.48 ± 2.15 μg mL −1 in assay effective capacity IC 4.28 0.19 mg . total content 662.51 20.95 gallic acid equivalents g , 114.99 0.51 quercetin demonstrating extract's richness Significant antiproliferative effects observed at dose 250 Inflammatory indicated reduction pro‐inflammatory responses. showed binding affinities between suggesting mechanisms action. Abstract Synthesis of novel unnatural amino acids (UAAs) from 4‐oxo‐4‐phenylbut‐2‐enoic acid derivatives with intramolecular aza‐Michael addition reaction in the presence chlorosulfonyl isocyanate (CSI) was reported soft conditions without any metal catalyst. Acids and base as a catalyst, solvents effects were investigated for synthesis UAAs. This method provides inexpensive, practicable, efficient approach to generate The use UAAs has attracted great interest development therapeutic agents drug discovery improve their properties. In this context, new UAAs, inhibition on important metabolic enzymes acetylcholinesterase (AChE) carbonic anhydrases I II (hCA II) investigated. compound 2g showed best CA AChE enzymes, while 2i exhibited profile against isoenzyme. values these compounds found 1.85 ± 0.64 AChE, 0.53 0.07 hCA I, 0.44 0.15 µM II, respectively, they stronger inhibitory property than acetazolamide (standard inhibitor tacrine AChE) molecules. activity studied molecule different proteins that are (PDB ID: 2CAB), 5AML), 1OCE) examined. Finally, properties examined by performing absorption, distribution, metabolism, excretion, toxicity analysis.

Abstract Objectives Melanoma is a highly malignant and serious form of skin cancer. In addition to the standard treatments, complementary approaches, including phytotherapy, are also used alleviate symptoms improve patient well-being. This study aims investigate anticancer effects Gypsophila eriocalyx (GE), an endemic species from Türkiye, on melanoma cells. We set out determine efficacy GE in inhibiting cell proliferation, migration, growth, explore its underlying mechanisms. Methods examined impact proliferation two lines, Malme-3M SK-MEL-28, assessed developmental toxicity zebrafish embryos. Next, we evaluated GE’s influence colony formation wound healing cells, as well ability induce apoptosis affect TGF-β/Smad signaling pathway, by measuring pathway reporter activity target gene expression. Results inhibited lines at concentrations 104 488 times lower than those required for normal non-malignant L929 fibroblast embryos, demonstrated only above 50 μg/mL. treatment significantly impaired abilities indicating reduced migration. Moreover, induced cells evidenced decreased Conclusions highlights potential novel therapeutic agent demonstrating inhibit tumor growth progression.

Abstract Introduction Osteoporosis, one of the common bone diseases, manifests itself as a decrease in mass. Recently, use medicinal plants search for effective and low‐toxicity therapeutics prevention or treatment osteoporosis has become trending topic. Objective In this study, we aim to prepare controlled drug carrier system loaded with Gypsophila eriocalyx determine its potential anti‐osteoporosis applications. Methods extract (GEE) was prepared, components were determined. The molecular interactions Cathepsin K (CatK), which is used target development against osteoporosis, revealed by silico docking MD methods. ADMET profiles also examined. GEE‐loaded chitosan nanoparticles (CNPs) synthesized. nanoparticles' morphology, encapsulation efficiency, loading capacity, release profile, average size, polydispersity index, zeta potentials cytotoxic effects GEE CNPs on L929 osteogenic proliferation human marrow stem cells (hBMC) Results analysis no breaks atomic changes dynamic system, confirmed continued interaction identical residues. It determined that CNP formulation produced successfully, had toxic effect L929, an hBMC. Conclusion line vitro results obtained, it evaluated can be candidate phytotherapeutic properties treatment.q1
