ABSTRACT Keloid scarring is a complex fibroproliferative disorder characterised by excessive fibroblast proliferation. Inhibition of cellular glycolysis effectively suppresses the proliferation keloid fibroblasts (KFs). Neural precursor cell‐expressed developmentally downregulated gene 4‐like (NEDD4L), ubiquitin ligase, regulates cell in different diseases. This study investigated effects NEDD4L on glucose metabolism, and migration KFs. Primary KFs were isolated from skin tissues obtained patients with active‐stage keloids. Cell transfection was used to upregulate or downregulate Yin Yang 1 (YY1) Protein expression assessed immunohistochemistry western blotting. Objective This study aims to screen common immunological markers of lung tissues and blood for diagnosis tuberculosis (TB). Methods Differentially expressed miRNAs (DEmRs) mRNAs (DEGs) were obtained by whole-transcriptome sequencing profiles on 18F-FDG PET/CT high low metabolic active regions in nine TB patients. Common screened intersecting with DEmRs, four miRNA GEO datasets, their target predicted through the miRTarbase Tarbase databases. Then these intersected DEGs, from samples immune-related genes, construct a miRNA-mRNA interaction network, hub genes identified Cytoscape. The relationship between immune infiltration evaluated using Cibersort. Finally, diagnostic model based Lasso regression analysis was established validated qRT-PCR. Results Five both tissues. Abstract Purpose Ubiquitination plays a crucial role in various diseases. This study aims to explore the potential ubiquitination related genes IPF. Methods The gene microarray dataset GSE24206 was obtained from GEO database. Subsequently, through differential expression analysis and molecular signatures database, we 1734 differentially expressed 742 genes. Through venn diagram analysis, 53 Then, gene-ontology (GO) enrichment Kyoto Encyclopedia of Genes Genomes (KEGG) pathway protein-protein interactions (PPI) set (GSEA) were applied for Finally, CDC20 ITCH IPF patients cells validated by qPCR western blot assay. Results A total (36 up-regulated 17 down-regulated genes) identified between 6 healthy controls. GO KEGG mainly involved regulation protein ubiquitination, post-translational modification ubiquitin mediated proteolysis. PPI results demonstrated that these interacted with each other. GSEA some hub epithelial mesenchymal transition, inflammatory response, hypoxia, apoptosis. experiment level consistent bioinformatics results. Hermansky–Pudlak syndrome (HPS)-associated pulmonary fibrosis (HPS-PF) is a progressive lung disease that major cause of morbidity and mortality in HPS patients. Previous studies have demonstrated the proteins play an essential role biogenesis function lysosome-related organelles (LROs) alveolar epithelial type II (AT2) cells found HPS-PF associated with dysfunction AT2 abnormal immune reactions.
