Abstract BackgroundThe senescence of mesenchymal stem cells (MSCs) greatly compromises their therapeutic effect in regenerative medicine. Numerous studies are focusing on possible rejuvenation strategies to enhance the efficacy autologous MSC-based therapy. S-adenosyl-L-methionine (SAM) is a metabolite present all living cells. However, there little research about effects and mechanisms SAM MSCs.MethodsIn this study, adipose-derived MSC (ADSC) were assessed vitro by β-galactosidase staining, reactive oxygen species assay, cell cycle tests, Western blot. The osteogenic adipogenic differentiation ability changes also detected. Mouse models premature aging established subcutaneous injection D-Galactose (D-gal). vivo antiaging roles tested through behavioral organ coefficient, pathological morphology, expression aging-related proteins major organs, such as heart, liver, kidney. bone density distal femur, well volume, number, thickness trabecula evaluated micro-CT. molecular searched validated transcriptome sequencing, blot, immunofluorescence. Transcriptome sequencing results revealed that PI3K/AKT involved SAM-mediated anti-senescence MSCs. Mechanistically, activated signaling increased phosphorylation FOXO3a, resulting decrease translocation FOXO3a nucleus inhibition activity P21. Ultimately, attenuated ADSCs potential.ConclusionsIn summary, our suggested could ameliorate MSCs PI3K/AKT/FOXO3a signaling. Meanwhile, exert vivo.

Abstract Luteolin is an essential natural polyphenol found in a variety of plants. Numerous studies have supported its protective role neurodegenerative diseases, yet the research for therapeutic utility D-galactose (D-gal)-induced brain ageing still lacking. In this study, potential neuroprotective impact luteolin against D-gal-induced was explored. Forty rats were randomly divided into four groups: control, luteolin, D-gal, and luteolin-administered D-gal groups. All groups subjected to behavioural, cholinergic function, hippocampal mitochondrial respiration assessments. Hippocampal oxidative, neuro-inflammatory, senescence apoptotic indicators detected. Gene expressions SIRT1, BDNF, RAGE assessed. histopathological studies, along with GFAP Ki67 immunoreactivity, performed. Our results demonstrated that effectively alleviated cognitive impairment reversed abnormalities. Furthermore, administration substantially mitigated hippocampus oxidative stress, dysfunction, neuro-inflammation, triggered by D-gal. Additionally, treatment considerably attenuated neuronal apoptosis upregulated SIRT1 mRNA expression. conclusion, our findings revealed D-gal-evoked senescence, improving function enhancing neuroregeneration rat model through antioxidant, senolytic, anti-inflammatory, anti-apoptotic impacts, possibly due upregulation SIRT1. could be promising modality aging-associated

Hyperbaric oxygen therapy (HBOT), a technique through which 100% is provided at pressure higher than 1 atm absolute (ATA), has become well-established treatment modality for multiple conditions. The noninvasive nature, favorable safety profile, and common clinical application of HBOT make it competitive candidate several new indications, one them being aging age-related diseases. In fact, despite the conventional wisdom that excessive accelerates aging, appropriate protocols without exceeding toxicity threshold have shown great promise in therapies against aging. Probiotics and prebiotics have received attention in alleviating neurodegenerative diseases. Lactobacillus plantarum (L. plantarum) 69-2 was combined with galactooligosaccharides (GOS) supplemented a d-galactose (d-gal)-induced neurodegeneration memory impairment mice model to explore its effects on the brain regulation of short-chain fatty acids. The results showed that L. plantarum-GOS supplementation inhibited d-gal-induced oxidative stress increased brain's nuclear factor erythroid 2-related 2 (Nrf2) levels. Butyrate, metabolite gut microbiota regulated by GOS, inhibits p-JNK expression, downregulates pro-apoptotic proteins expression activation inflammatory mediators, upregulates synaptic protein expression. This might be potential mechanism for GOS alleviate impairment. study sheds new light development aging-related neuroprotective dietary supplements based gut–brain axis.

Abstract Bisphenol A (BPA) in the environment can have deleterious effects on humans and animals. BPA exert nephrotoxicity by inducing oxidative stress. Selenium (Se) deficiency specifically impair kidney tissues additionally show a synergistic effect toxicity of several environmental chemicals. However, toxic chicken whether Se produces remain poorly understood. Herein, we established exposure models model vivo vitro, described discovery path aggravation apoptosis necroptosis Se‐deficient kidneys via regulation stress phosphatidylinositol 3‐kinase/threonine kinase (PI3K/AKT) signaling pathway. We found that increased reactive oxygen species malondialdehyde levels, reduced activities catalase, GPx, superoxide dismutase, downregulated PI3K AKT expressions, activated Bcl/Bax‐Caspase 9‐Caspase 3, receptor‐interacting protein 1/mixed lineage domain‐like pathways, resulting kidney. In addition, significantly promoted expression renal BPA‐exposed kidneys. Altogether, our results showed aggravates PI3K/AKT Our findings elucidate mechanism exacerbation chickens will provide great significance for protection ecological animal health.

Skin problems caused by aging have attracted much attention, and marine collagen peptides been proved to improve these problems, while mammalian are rarely reported. In this study, fermented deer bone peptide (FCP) non-fermented (NCP) were extracted from bone, respectively, their sequences differential proteins analyzed using LC-MS/MS technology. After they applied mice induced with D-gal, the skin hydration ability, antioxidant synthesis, degradation ability of studied. The results show that FCP NCP mainly constitute type Ⅰ collagen, segments different. vivo experiments can richness fibers in mice; skin; promote activity antioxidant-related enzymes; also through TGF-β MAPK pathways, synthesis regulated. Abstract Background Hydrogen sulfide (H 2 S), the third gasotransmitter discovered, regulates a variety of physiological functions. Whether H S alleviates skeletal muscle ageing by regulating autophagy has not been reported. Methods Mice were administered 150 mg/kg/day D ‐galactose ( ‐gal), and C2C12 myotubes cultured in 20 g/L ‐gal to induce ageing. Sodium hydrosulfide (NaHS) was employed as an exogenous donor treatment group. The intracellular concentration quantified 7‐azido‐4‐methylcoumarin fluorescence probe. proteins involved ubiquitin‐mediated degradation AMPKα1 detected liquid chromatography tandem mass spectrometry (LC–MS/MS) co‐immunoprecipitation (Co‐IP). Previous reports have confirmed that saponins (ginsenosides) derived from Panax ginseng. C. A. Meyer exerted obvious memory-enhancing and antiaging effects, the simpler structure of ginsenosides, better biological activity. In this work, we aimed to explore therapeutic effect underlying molecular mechanism 20(S)-protopanaxatriol (PPT), aglycone panaxatriol-type by establishing D-galactose (D-gal)-induced subacute brain aging model in mice. The results showed PPT treatment (10 20 mg/kg) for 4 weeks could significantly restore D-gal (800 mg/kg 8 weeks)-induced impaired memory function, choline dysfunction, redox system imbalance Meanwhile, also reduced histopathological changes caused exposure. Moreover, increase TFEB/LAMP2 protein expression promote mitochondrial autophagic flow. Importantly, docking had good binding ability with LAMP2 TFEB, suggesting might play an important role alleviate D-gal-caused aging.
