Atopic dermatitis (AD), a chronic skin condition characterized by itching, redness, and inflammation, is closely associated with heightened levels of endogenous reactive oxygen species (ROS) in the skin. ROS can contribute to onset progression AD through oxidative stress, which leads release proinflammatory cytokines, T-cell differentiation, exacerbation symptoms. In this study, we aim develop therapeutic antioxidant hydrogel patch for potential treatment using lignin, biomass waste material. Lignin contains polyphenol groups that enable it scavenge exhibit properties. The lignin patches, possessing optimized mechanical properties control cross-linker ratio, demonstrated high ROS-scavenging capabilities. Furthermore, excellent biocompatibility skin, exhibiting beneficial protecting human keratinocytes under conditions. When applied an mouse model, effectively reduced epidermal thickness inflamed regions, decreased mast cell infiltration, regulated inflammatory cytokine levels. These findings collectively suggest serves as managing modulating stress its ability.

Cutaneous eczema is a kind of skin disease characterized by inflammation. The main manifestations are various types dermatitis, eczema, and urticaria. There usually complications such as erythema, blisters, epidermal peeling. quercetin might have therapeutic effect on cutaneous due to its favorable antioxidant activity anti-inflammatory effects. Currently, there few studies transdermal administration drugs for the treatment eczema. aim this study was prepare quercetin-containing liposomes-in-gel (QU-LG), properties were evaluated, it used in mice suffering from dermal see if had preventive effects an attempt make new option QU-LG prepared injection method form liposomes (QU-L) evenly dispersed natural dissolution carboxymethylcellulose sodium (1%, CMC-Na). release across dialysis membranes up 30% clearance 1,1-diphenyl-2-picrylhydrazyl (DPPH) 65.16 ± 3.513%. In anti-oxidation assay inhibited malondialdehyde (MDA) production liver better than commercially available drug dexamethasone acetate cream. Compared with untreated mice, treated showed statistically significant reduction dermatopathologic symptoms. results suggested that good vivo vitro could be prevention

Atopic dermatitis (AD) is the most common chronically relapsing inflammatory skin disease, predominantly in children; it characterized by an eczematous pattern generally referable to dryness and itchy papules that become excoriated lichenified more advanced stages of disease. Although pathophysiology AD not completely understood, numerous studies have demonstrated complex interaction between genetic, immunological, environmental factors, which acts disrupt barrier function. Free radicals play a key role directly damaging structure, inducing inflammation weakening barrier. Tempol (4-hydroxy-2,2,6,6-tetramethylpiperidine-1-oxyl) membrane-permeable radical scavenger, known be stable nitroxide, exhibits excellent antioxidant effects several human disorders, such as osteoarthritis bowel diseases. Considering few existing on dermatological pathologies, this study aimed evaluate tempol, cream formulation, murine model AD. Dermatitis was induced mice via dorsal application 0.5% Oxazolone, three times week for two weeks. After induction, were treated with tempol-based another weeks at different doses 0.5%, 1% 2%. Our results ability highest percentages, counteract reducing histological damage, decreasing mast cell infiltration, improving properties, restoring tight junction (TJs) filaggrin. Moreover, 2%, able modulate nuclear factor kappa-light-chain-enhancer activated B (NF-κB) pathway, well tumor necrosis (TNF)-α interleukin (IL)-1β expression. Topical treatment also attenuated oxidative stress modulating erythroid 2-related 2 (Nrf2), manganese superoxide dismutase (MnSOD), heme oxygenase I (HO-1) expression levels. The obtained demonstrate advantages provided topical administration through modulation NF-κB/Nrf2 signaling pathways. Therefore, tempol could represent alternative anti-atopic approach treating AD, thereby

Atopic dermatitis (AD) is a common inflammatory of the skin and poses therapeutic challenges due to adverse reactions high costs associated with available treatments. In Eastern Asian countries, plethora herbal remedies extensively employed for alleviation AD. Many these botanicals are renowned their formidable anti-inflammatory properties, contributing AD management. Chinese medicine (CHM) its active ingredients exhibit both prophylactic promise against by modulating response, orchestrating immune system functions, enhancing antioxidant activities. A comprehensive exploration underlying mechanisms involved in CHM treatment can enhance comprehension pathogenesis facilitate development innovative drugs This study aims elucidate signaling pathways potential targets implicated CHM-based AD, providing systematic theoretical framework application therapy while serving as valuable reference developing more effective safer agents.

Atopic Dermatitis (AD) is a common continuous inflammation dermatosis requiring efficacious therapeutic intervention. Phellodendri Chinensis Cortex-Cnidii Fructus (PC) herb pair has shown effectiveness and security in traditional Chinese medicine (TCM) clinical applications, yet its pharmacological constituents mechanisms are not fully elucidated. This study used serum pharmacochemistry, network pharmacology, validation experiments to examine the impact of PC treatment AD. Initially, ultra performance liquid chromatography-mass spectrometry (UPLC-MS) had been applied elucidate components that were absorbed. An integrative approach combining pharmacology vivo research (general index observation, skin pathological tissue staining, ELISA, immunohistochemistry, immunofluorescence, Western blotting) was employed validate PC's mechanism action after 2,4-dinitrochlorobenzene (DNCB) create mouse model Fifty-three compounds 18 prototype characterized within PC. The efficacy AD notably manifested alleviation pruritus, improvement histopathology, reduction cytokines involving IgE, IL-4, TNF-α IL-6. Based on molecular docking studies, pharmacodynamic such as phellodendrine, xanthotoxin, nomilin, isopimpinellin strongly favored main targets. Comprehensive investigations integrating studies revealed prevented DNCB-induced through adjusting TLR4/NF-κB signaling pathway. anti-AD effects may be attributed modulation pathway, NF-кB expression nucleusim, downregulation inflammatory cytokine levels, provement histopathological manifestations, pruritus.

Abstract The skin is the body's largest organ and performs several vital functions, such as controlling movement of essential substances while protecting against external threats. Although mainly composed keratinocytes (KCs), also contains a complex network immune cells that play critical role in host defense maintaining homeostasis. KCs proliferate basal layer epidermis undergo differentiation, altering their functional phenotypic characteristics. These differentiation steps are crucial for stratification formation stratum corneum, ensuring barrier's functions. Exposure to UV, environmental pollutants, or chemicals can lead an overproduction reactive species oxygen (ROS), leading oxidative stress. To ensure redox homeostasis prevent damage resulting from ROS, has extensive antioxidant systems, orchestrated by nuclear factor erythroid‐2‐related 2 (Nrf2) pathway. Indeed, Nrf2 induces expression detoxification enzymes suppresses inductions pro‐inflammatory cytokine genes. In this context, preserving functions epidermal regulating immunity, managing stresses. Besides, pathway plays important pathogenesis common inflammatory diseases allergic contact dermatitis, atopic psoriasis. Therefore, present review highlights well its contribution pathophysiology diseases. Finally, particular emphasis will be placed on therapeutic potential targeting alleviate symptoms these disorders.
