Drought and metal pollution severely impact plant growth. Root-associated extremophilic fungi can improve performance, their encapsulation improves protection effectiveness. This study optimized the conditions for an fungus with growth-promoting traits using alginate-chitosan capsules. An endophytic was isolated from roots of Neltuma chilensis Atacama Desert identified via internal transcribed spacer (ITS) sequencing. Its traits, including exopolysaccharide, ammonium, siderophore, indole acetic acid production phosphorus solubilization, were evaluated. Freeze-dried Penicillium nalgiovense encapsulated jet-breaking extrusion, capsule morphology fungal survival assessed scanning electron microscope (SEM), confocal laser microscopy (CLSM), viability tests. Using Taguchi's design, optimal sphericity (0.914 ± 0.002) mean size (3.232 0.087 mm) achieved 1% chitosan, a 5 cm distance to gelation bath, 40 Hz vibration frequency. CLSM analysis confirmed presence chitosan outer layer, revealing capsule's coating material encapsulating P. nalgiovense. The remained viable across disinfection times, demonstrating effective gradual release. These findings emphasize need precise parameter control in encapsulation, providing basis developing robust bioinoculants support resilience extreme environments.
